摘 要:采用一步水热法制备具有类过氧化物酶活性的MoS_(2)纳米片,并基于MoS_(2)纳米片可催化H_(2)O_(2)氧化3,3′,5,5′-四甲基联苯胺(TMB)生成蓝色氧化产物(ox-TMB)的原理,建立了测定H_(2)O_(2)浓度的分析方法。在pH 3.7、温度35℃、MoS_(2)浓度1.0μg/mL及孵育时间40 min的条件下,ox-TMB的吸光度随H_(2)O_(2)浓度的升高而增大。基于H_(2)O_(2)是尿酸氧化酶催化尿酸氧化的产物之一,建立了MoS_(2)纳米片催化H_(2)O_(2)氧化TMB间接测定尿酸的分析方法。在2.5~40μmol/L和40~100μmol/L范围内,检测体系的吸光度与尿酸浓度呈良好的分段线性关系,方法在低浓度区间的检出限为0.093μmol/L,应用于血清和尿样中尿酸的测定,回收率为92.6%~102.0%。MoS_(2)nanosheets with peroxidase-like activity were prepared by one-step hydrothermal method.The obtained MoS_(2)nanosheets could catalyze H_(2)O_(2)-mediated oxidation of 3,3′,5,5′-tetramethylbenzidine(TMB)to produce corresponding blue oxidized product(ox-TMB).Thus,a simple method for detection of H_(2)O_(2) was established.Under the optimized conditions such as pH 3.7,temperature of 35℃,1.0μg/mL MoS_(2)and incubation time of 40 min,the absorbance of ox-TMB increased with the increase of H_(2)O_(2) concentration.Given the fact that H_(2)O_(2) was one of the products of uric acid oxidase-catalyzed uric acid oxidation,a method for the indirect detection of uric acid based on MoS_(2)nanosheets-catalyzed and H_(2)O_(2)-mediated TMB oxidation was developed.The increase of ox-TMB absorbance enabled the sensitive determination of uric acid in the lineatrange of 2.5-40μmol/L and 40-100μmol/L,respectively,and the detection limit was 0.093μmol/L(3σ/k).The method exhibited potential application in the detection of uric acid in urine and serum samples with recoveries of 92.6%-102.0%.
