摘  要:环境二噁英类污染物受到广泛关注,目前常用的二噁英类物质检测方法有仪器分析法(高分辨率气相色谱-高分辨率质谱法)和生物检测法。生物检测法具有便捷高效、成本低廉等优点,但其测定值普遍高于仪器分析法,关键原因是生物检测法在前处理阶段未能完全消除非二噁英类物质的干扰。改进了传统7-乙氧基-3-异吩恶唑酮-脱乙基酶(EROD)生物检测法的前处理方法,并利用改进后的EROD法与仪器分析法对相同环境样品进行检测,对比分析两种方法得到的二噁英毒性当量数据。结果显示,改进后的细分前处理手段结合梨形烧瓶-Mix前处理手段能有效提高二噁英类物质的回收率(接近100%),并且改进后的EROD法的检测结果与仪器分析法测定的二噁英当量结果相当,说明通过改进前处理方法可以将生物检测法的准确度提高到与仪器分析法一致的水平。研究结论可为生物检测法在环境样品二噁英类污染物检测中的应用和推广提供有效的数据参考。Dioxins in the environment have been drawn extensive attention.Currently,the commonly used detection methods for dioxins include high resolution gas chromatography-high resolution mass spectrometry(HRGC-HRMS)and bioassay methods.Bioassay method has the advantages of convenience,high efficiency and low cost,but the analysis results of bioassay method are higher than that of HRGC-HRMS.This is because the non-dioxins in the pretreatment processes in the bioassay method cannot be completely removed.In this study,the pretreatment processes in the bioassay method were improved,and the comparison analysis results indicated that the improved methods including the subdividing+pear-shaped flask-Mix can in which EROD enzyme activity induced method is a commonly used and highly effective bioassay method for the detection of dioxins.This paper mainly introduces a sample preparation method for improving the recovery rate of dioxins in EROD bioassay method,which can efficiently recover dioxin substances dissolved in solvents.The toxic equivalent quantity of dioxins in samples determined with the improved pretreatment methods in the bioassay methods were equal to that determined with HRGC-HRMS methods,indicating that the improved pretreatment methods can enhance the detection sensitivity of dioxins in the bioassay methods.This study provides the effective data support for the application of bioassay method in the detection of dioxins in environmental samples.
