基于网络药理学探讨中药调控肿瘤相关性细胞胀亡用药规律

摘　　要：目的探讨中药调控肿瘤相关性细胞胀亡的物质基础及用药规律。方法通过检索KEGG、GeneCards、OMIM数据库获取细胞胀亡的作用靶点,以类药性≥0.18为条件,通过中药系统药理学数据库与分析平台(TCMSP)筛选出可作用于靶点的化合物及中药,并构建靶点-化合物、化合物-中药、靶点-化合物-中药网络。依据2015年版《中华人民共和国药典》及《中药学》确定药物的性味归经,分析用药规律。结果共获得34个可作用于细胞胀亡的靶点,可匹配到候选化合物的靶点11个,得到70个候选化合物和324味中药。可干预细胞胀亡的药味以苦、甘、辛为主,药性以寒居多,主要归肝、肺、胃经。结论本研究较系统地探讨了中药干预肿瘤相关性细胞胀亡的物质基础,总结了相关中药的一般规律,可为中西医结合临床及干预细胞胀亡相关中药新药研发提供思路和依据。Objective To investigate the material basis and medication laws of Chinese materia medica in regulating tumor-associated oncosis. Methods The targets of oncosis were obtained by searching the KEGG, GeneCards, and OMIM. On the condition of drug-like property(DL) ≥ 0.18, compounds and Chinese materia medica acting on the targets were selected from the TCMSP, and the target-compound, compound-Chinese materia medica, target-compound-Chinese materia medica networks were constructed. According to Chinese Pharmacopoeia and Chinese Materia Medica, property, taste and meridian tropism of each Chinese materia medica were identified, and the medication laws were analyzed. Results A total of 34 targets for oncosis were obtained, among which 11 targets were matched to compounds. 70 candidate compounds and 324 kinds of Chinese materia medica were obtained. The results of frequency statistics showed that bitter, sweet and pungent were the main properties which could interfere oncosis, the tastes were mainly cold, and the meridian tropism belonged to liver, lung and stomach. Conclusion This study systematically explores the material basis of TCM intervention in tumor-associated oncosis, summarizes the general rules of Chinese materia medica, and provides ideas and basis for the future clinical application of integrated traditional Chinese and Western medicine and the development of new TCM medicine for intervention in cell oncosis.
