摘  要:为了便于对我国三带喙库蚊种群的遗传结构和多样性进行动态检测,本研究利用磁珠富集法进行三带喙库蚊微卫星标记的开发和筛选,共获得40个微卫星位点,且位点的多态性验证结果表明:有24个位点在不同三带喙库蚊地理株间呈现多态性。微卫星位点的遗传多样性分析结果表明:各位点的平均多态信息含量(PIC)和香农指数(SI)均较高,其中位点P17最高(PIC=0.853,SI=2.281),最低的为位点P40(PIC=0.503,SI=1.195),都属于高度多态性位点,适用于后续三带喙库蚊种群遗传多样性及遗传结构的研究。To monitor the dynamic changes of genetic diversity and population structure of Culex tritaeniorhynchus populations in China,a total of 40 microsatellite loci were developed from its genome via Magnetic Beads Enrichment method.The polymorphism verification result showed that 24 loci of all displayed polymorphic among Cx.tritaeniorhynchus populations from different geographic regions.Meanwhile,genetic diversity analysis results of these microsatellite loci showed that both of the average polymorphic information content(PIC)and Shannon index(SI)of these loci were high,with the highest one was locus P17(PIC=0.853,SI=2.281)and lowest one was locus P40(PIC=0.503,SI=1.195).Therefore,these new developed microsatellite loci are suitable for the subsequent study of the genetic diversity and population structure of Cx.tritaeniorhynchus.
