摘 要:目的:对田基黄饮片质量标准进行研究。方法:采用显微、TLC对田基黄进行定性鉴别;用中国药典法对田基黄的水分、总灰分、酸不溶性灰分和浸出物进行测定;采用HPLC法测定槲皮苷和异槲皮苷含量,色谱柱为月旭-C18柱(250 mm×4.6 mm,5μm);流动相:乙腈-2.5%冰醋酸(19∶81);流速:1.0 ml·min^(-1);柱温:30℃;检测波长:355 nm。结果:显微特征明显;薄层斑点清晰,分离度好;槲皮苷、异槲皮苷分别在10.77~269.29μg·ml^(-1)、5.58~139.58μg·ml^(-1)范围内线性关系良好(r=0.999 9),平均加样回收率分别为98.22%,98.63%,RSD分别为1.31%,1.54%(n=6)。结论:本法简单易行,可操作性强,可用于田基黄饮片质量控制。Objective: To study the quality standard for Hypericum japonicum. Methods: Microscopy and TLC were used to identify the characteristics of Hypericum japonicum. The contents of water,total ash,acid-insoluble ash and ethanol soluble extracts were determined according to the methods described in Chinese Pharmacopoeia. HPLC was used for the content determination of quercitrin and isoquercitrin. A Wech-C18( 250 mm×4.6 mm,5 μm) column was used as the analytical column,the mobile phase was acetonitrile-2.5%acetic acid( 19∶81) at the flow rate of 1.0 ml·min^(-1),the temperature was 30℃ and the detection wavelength was 355 nm.Results:Microscopic characteristics were obvious;TLC showed clear spots and good resolution. Quercitrin and isoquercitrin showed good linear relationship within the range of 10.77-269.29 μg·ml^(-1) and 5.58-139.58 μg·ml^(-1)( r = 0.999 9),and the average recovery was 98.22% and 98.63% with the RSDs of 1.31% and 1.54%( n = 6),respectively. Conclusion: The method is simple and easy to operate,and can be used for the quality control of Hypericum japonicum.
