龙蒲定神丸对小鼠行为学的影响及其急性毒理学研究

摘　　要：目的:观察龙蒲定神丸(DSW)对小鼠行为学的影响并考察其急性毒性。方法:将60只小鼠随机分为5组,每组12只,阴性对照组(生理盐水)、阳性对照组(养血安神片2.36g·kg-1)、DSW高剂量组(5.71g·kg-1)、中剂量组(2.86g·kg-1)、低剂量组(1.43g·kg-1),灌胃给药。通过小鼠的自主活动实验、避暗实验以及戊巴比妥钠催眠实验,观察小鼠运动行为的变化;按改良寇氏法测定其LD 50,评价DSW的安全性。结果:DSW高剂量组小鼠自主活动量和站立次数明显降低,DSW低剂量组小鼠站立次数明显减少;DSW高、中剂量组小鼠犯错误次数明显减少,DSW中、低剂量组小鼠逃避潜伏期明显缩短;DSW高、中剂量组小鼠入睡率明显增加,DSW各剂量组小鼠入睡潜伏期明显缩短。DSW给药剂量2.86g·kg-1、11.44g·kg-1、34.32g·kg-1,均未见动物死亡。结论:DSW能改善小鼠的自主活动、增强学习记忆,并具有镇静催眠作用;DSW的毒性甚小,在规定剂量范围内应用是安全可靠的。Objective:To evaluate the effect of Long Pu Ding Shen Wan(DSW)on the behavior of mice and the safety of DSW.Methods:A total of 60 mice were randomly divided into 5 groups with 12 each,negative control group(normal saline),positive control group(Yang’xue anshen pian,2.36 g·kg-1),high-dose group(DSW,5.71g·kg-1),medium-dose group(DSW,2.86 g·kg-1),low-dose group(DSW,2.86 g·kg-1),intragastric administration.The changes in the movement behavior of the mice were observed through spontaneous activity experiment,step-through task and hypnotize experiment induced by pentobarbital sodium.The safety of DSW was evaluated by the modified Koch method.Results:the spontaneous activity and standing times of the mice in the high-dose group were significantly decreased,and the standing times of the mice in the low-dose group were significantly decreased;the number of errors in the high-dose and medium-dose groups was significantly reduced,and the escape period in the low-dose and medium-dose groups was significantly shortened.The sleep rate of mice in the high and medium dose groups was significantly increased,and the escape period in each dose group was significantly shortened.The dosages of DSW were 2.86 g·kg-1,11.44 g·kg-1 and 34.32 g·kg-1.No animals died.Conclusion:DSW can improve autonomic activity,improve learning and memory,and have sedative and hypnotic effect.The toxicity of DSW is very small,and it is safe and reliable to use within the prescribed dose range.
