摘  要:重金属转运P1B型ATP酶对促进金属离子的吸收、运输、积累、分配和解毒至关重要。其中HMA3蛋白可能通过将镉螯合至液泡从而有助于金属解毒,本研究以不同浓度Cd处理生物质高粱幼苗为材料,克隆获得了SoHMA3基因的全长序列,cDNA全长3179 bp,编码877个氨基酸,编码蛋白的理论分子量为94.44 kD,理论等电点为6.4,为跨膜蛋白,该蛋白属于HAD超家族成员,对该氨基酸序列进行聚类分析表明,其与玉米和水稻的同源性最高,而与芦笋最低。实时定量PCR分析表明,SoHMA3基因在生物质高粱地上部和地下部均有表达,不存在组织特异性;镉胁迫下,该基因在地上部无显著响应,在地下部有显著响应,表明生物质高粱SoHMA3的表达受到Cd胁迫的影响,参与了生物质高粱对Cd的解毒。本研究结果为进一步探究生物质高粱对Cd的吸收、转运及解毒机理提供基础资料。Heavy metal transporter P1 B atpase plays an important role in promoting the absorption,transportation,accumulation,distribution and detoxification of metal ions,HMA3 may detoxify the metal by chelating cadmium into vacuoles.In this study,biomass sorghum seedlings were treated with different concentrations of Cd,and the full-length sequence of SoHMA3 gene was cloned which was 3179 bp,encoding 877 amino acids,with a predicted molecular mass of 94.44 k D and a basic isoelectric point of 6.4.It is a transmembrane protein belonging to the HAD superfamily.Phylogenetic tree analysis showed that the protein was most closely related to corn and rice.Real-time quantitative PCR analysis showed that So HMA3 gene was expressed in both the abovematic and subterranean parts of biomass sorghum.Under cadmium stress,it had no significant response in the aboveground part,but had an obvious response in the underground part,indicating that the expression of SoHMA3 in biomass sorghum was affected by Cd stress and participated in the detoxification of Cd to biomass sorghum.The results of this study provide basic data for further exploring the mechanism of Cd absorption,transport and detoxification of biomass sorghum.
