摘 要:WRKY基因家族是一类含有WRKY保守结构域的转录因子大家族,是植物中最大的转录调控因子家族之一。目前已有多种植物WRKY家族的分析鉴定报告,但关于甘薯(Ipomoea batatas(L.)Lam.)中WRKY家族的报告却很少。本研究利用生物信息学方法对甘薯近缘野生种三浅裂野牵牛Ipomoea trifida(Kunth)G.Don的基因组数据进行了详细的分析,结果显示:在三浅裂野牵牛基因组中共鉴定得到82个WRKY转录因子,其中81个不均匀分布在基因组的15条染色体上,第5号染色体上分布最多(10个基因),第8和15号染色体上分布最少(2个基因);蛋白分子大小在116~710个氨基酸之间,等电点为4.85~10.33不等;按照保守结构域分类可以分为Ⅰ、Ⅱ、Ⅲ三组,Ⅱ组包含Ⅱa、Ⅱb、Ⅱc、Ⅱd和Ⅱe五个亚组,其中Ⅰ组有15个ItWRKY基因、Ⅱa组4个、Ⅱb组11个、Ⅱc组23个、Ⅱd组7个Ⅱe组10个、Ⅲ组12个;82个ItWRKY基因中有8个基因的核心结构域WRKYGQK发生了单个氨基酸变化(WRKYGKK)。ItWRKY基因以组织特异性的方式表达且在不同胁迫下基因的表达量存在差异,表明ItWRKY基因参与了三浅裂野牵牛对胁迫的响应。本研究为甘薯WRKY基因的功能鉴定提供了参考作用,进一步为甘薯的分子育种提供了帮助。WRKY is a superfamily of transcription factor,containing a conserved WRKY domain.They are of the largest family of transcription regulators in plants.WRKY genes have been identified with systematic analysis and comparison in various plants.However,little information has been reported about the systematic identification and analysis of WRKY genes in sweet potato.The genomic data of Sweet potato wild relative Ipomoea trifida were analyzed in detail using bioinformatics method in this study.The results showed that 82 ItWRKY genes were identified,81 WRKY genes with specific location information were distributed unevenly on all 15 chromosomes.Most of the WRKYs were presented on chromosome 5 H(10 WRKYs),while the fewest on chromosome 8 H and 15 H(2 WRKYs).ItWRKY proteins contained 116 to 710 amino acids and the isoelectric point ranged from 4.85 to10.33.To the structure of conserved domain,all together 82 WRKYs were classified into three major groups,including 15 typeⅠgenes,55 typeⅡgenes and 12 typeⅢgenes.Furthermore,typeⅡgenes can be divided into 5 groups,namelyⅡa(4),Ⅱb(11),Ⅱc(23),Ⅱd(7)andⅡe(10).ItWRKY proteins were found to have the highly conserved sequence WRKYGQK,while 8 ItWRKYs vary by a single amino acid(WRKYGKK).ItWRKY genes expressed with a tissue-specific manner and differently expressed under various abiotic stresses,suggesting that the ItWRKYs were involved in stress response in Ipomoea trifida.This study supplied a solid theoretical foundation for the functional study of WRKY gene family and further facilitated the molecular breeding of sweet potato.
