摘  要:为研究氯吡脲对葛根Ptexp1基因表达影响及其与产量和品质的相互关系,揭示氯吡脲对葛根产量和品质影响的分子机制,本研究以‘桂葛1号’粉葛幼叶为材料,采用PCR同源克隆和RACE技术,克隆得到葛根expansin基因,命名为Ptexp1(GenBank登录号:MK169414)。该基因cDNA全长1063 bp,开放阅读框为91~846 bp,编码251个氨基酸,预测其分子量和等电点分别为26.78 kD和8.37,在64~149 aa和160~237 aa有DPBB_1和Pollen_allerg_1两个保守的蛋白质结构域。该基因与其他植物的expansin-A8基因核苷酸序列和氨基酸序列高度同源,其中与大豆、绿豆和豇豆expansin-A8基因的核苷酸序列同源性分别高达94%、92%和91%,与鸡血藤、大豆、豇豆和相思子expansin-A8基因的氨基酸序列同源性分别高达97%、95%、94%和93%。以葛根18S作为内参基因,Ptexp1基因在葛根膨大期不同组织中的表达规律为块根>根蔸>叶>茎,与不同部位发育表型吻合。1 mg/L氯吡脲处理显著提高该基因在块根、根蔸和茎的表达,但对叶片表达的影响不显著,与其处理显著提高块根、根蔸和茎的形态指标相吻合。本研究为氯吡脲调控葛根膨大的作用机理、产量和品质形成的分子机制及其新品种选育提供参考。Effects of CPPU on the expression of Ptexp1 gene and its relationship with yield and quality formation were studied,to reveal the molecular mechanism on effects of CPPU in the yield and quality of Pueraria lobata.In the present study,a Ptexp1 gene encoding expansin was cloned from young leaves of’Gui-ge 1’using PCR homologous cloning and RACE technology.The Ptexp1 gene(GenBank accession number:MK169414)was 1063 bp in full length,and had an open reading frame in positions 91~846 bp,encoding 251 amino acids.The predicted molecular mass and isoelectric point were 26.78 k D and 8.37,respectively.Two conserved domains of DPBB_1 and Pollen_allerg_1 were shown in positions 64~149 aa and 160~237 aa,respectively.The gene was highly homologous to the nucleotide and amino acid sequences of the expansin-A8 gene in other plants,in which the nucleotide sequence homology was as high as 94%,92%and 91%with the ex pansin-A8 gene of Glycine max,Vigna radiata and Vigna unguiculata,respectively,in which the amino acid sequence homology was as high as97%,95%,94%and 93%with the expansin-A8 of Spatholobus suberectus,Glycine max,Vigna unguiculata and Abrus precatorius,respectively.With 18 S as the internal reference gene,the expression of Ptexp1 gene in different tissues of P.lobata during the root tuber expansion stage was as follows:root tuber>root hill>leaf>stem,which was consistent with the phenotypic development of different parts.The treatment of 1 mg/L CPPU significantly increased the expression of Ptexp1 in root tubers,root hills and stems,but the effects on the expression of leaves were not significant,which was consistent with the treatment that significantly improved the morphological indicators of root tubers,root hills and stems.This study provides references for the mechanism of CPPU in regulating expansion,the molecular mechanism of yield and quality formation and the breeding of new varieties in P.lobata.
