摘 要:目的探讨丹红注射液干预后对沙鼠前脑短暂性缺血再灌注的保护作用,及对热休克蛋白20(HSP20)和促红细胞生成素(EPO)表达的影响。方法将雄性蒙古沙鼠的双侧颈总动脉分离并夹闭10 min后恢复灌注血流建立前脑缺血再灌注模型。按照体重将沙鼠随机分为3组:假手术组,模型组和实验组,每组10只。实验组在造模前1天腹腔注射丹红注射液(8 mL·kg^(-1),qd)。造模后,分别于再灌注后第6 h以及第1,3和7天,用改良的神经功能缺损评分(MNSs)评价小鼠的运动功能,用免疫组织化学染色法检测HSP20和EPO水平(阳性细胞计数)。结果假手术组、模型组和实验组的MNSs分别为5.17±0.31,8.63±1.02和5.47±1.03;于第3天,这3组的HSP20表达量分别为4.70±0.91,95.10±2.18和118.2±1.61;这3组的EPO表达量分别为7.80±0.87,79.10±1.51和97.10±2.46。上述指标:模型组与假手术组相比,差异均有统计学意义(均P<0.01);实验组与模型组相比,差异均有统计学意义(P<0.05,P<0.01)。结论丹红注射液可能通过上调HSP20和EPO的表达,进而改善脑功能。Objective To observe the expression of heat shock proteins 20(HSP20)and erythropoietin(EPO)in brain tissue of Mongolian Gerbils after intervention of Danhong Injection with transient cerebral ischemia and reperfusion,and to explore the protective effect of Danhong Injection.Methods The anterior cerebral ischemia reperfusion model was established by isolating and trapping the bilateral common carotid arteries of healthy male Mongolian Gerbils for 10 min.The rats were randomly divided into 3 groups:sham operation group,model group and experimental group.Mongolian Gerbils in experimental group was intraperitoneally injected with Danhong Injection(8 mL·kg^(-1),qd)1 day before modeling,followed by ischemia reperfusion.The modified neurological severity scores(MNSs)were performed on 6 h,1 d,3 d and 7 d after cerebral ischemia reperfusion.The positive cell count of heat shock proteins 20(HSP20)and erythropoietin(EPO)were detected by immunohistochemical staining technique.Results The MNSs in sham operation group,model group and experimental group were 5.17±0.31,8.63±1.02 and 5.47±1.03,respectively;the levels of HSP20 at third in the three groups were 4.70±0.91,95.10±2.18,118.2±1.61,respectively;the levels of EPO at third in the three groups were 7.80±0.87,79.10±1.51,and 97.10±2.46,respectively.Compared between model group and sham operation group,the difference of the factors were significant(all P<0.01);compared between experimental group and model group,the difference of the factors were significant(P<0.05,P<0.01).Conclusion Danhong Injection may improve brain function by upregulation of HSP20 and EPO.
