含挥发油饮片标准汤剂探讨研究--以紫苏叶为例

摘　　要：目的建立紫苏叶标准汤剂质量评价方法,为紫苏叶标准汤剂质量标准的制定提供参考数据。方法按照含挥发油类饮片标准汤剂制备要求,制备17批紫苏叶饮片标准汤剂,考察紫苏叶标准汤剂挥发油转移率、出膏率及pH值,并建立紫苏叶标准汤剂HPLC指纹图谱分析方法。结果紫苏叶标准汤剂的出膏率范围为14.50%-24.80%,挥发油转移率范围为62.50%-100.00%;pH值范围为4.70-5.25。17批紫苏叶标准汤剂指纹图谱相似度均大于0.8,其中共有峰7个,指认3个并对这3个峰进行初步定量分析,探讨适合紫苏叶标准汤剂的定量指标成分。结论建立的紫苏叶标准汤剂质量评价方法可为紫苏叶标准汤剂质量标准的制定提供参考数据。在紫苏叶标准汤剂中,除了计算挥发油的转移率,亦可同时将迷迭香酸等作为其定量指标成分。Objective To establish the quality evaluation method for the standard decoction of Perillae Folium, and provide reference for the quality standard of standard decoction of Perillae Folium. Methods The standard decoction was prepared by using 17 batches of Perillae Folium. Then the transfer rate, dry extract rate and pH value were determined, and the analysis method of high-performance liquid chromatography(HPLC) fingerprint was established.Results The extraction rate was at the range of 14.50%-24.80%, the transfer rate ranged from 62.50% to 100.00%, and the pH value was at the range of 4.70-5.25. The similarity of fingerprint analysis for 17 batches of standard decoctions was more than 0.8. In total, 7 peaks were determined, in which 3 peaks were identified. The preliminary quantitative analysis was carried out on the 3 peaks and the index components for standard decoction of Perillae Folium were discussed. Conclusion The quality evaluation method for the standard decoction of Perillae Folium could provide reference in the establishment of quality standard for the standard decoction of Perillae Folium, and rosmarinic acid could also be used as a quantitative index component.
