摘 要:[目的]观察磁珠法在丙型肝炎病毒核酸定量检测中的应用效果.[方法]选择自2018年1月至2020年1月间采集的120例丙型肝炎患者的丙型肝炎病毒(HCV)血清标本,其中低病毒载量22例,中病毒载量64例,高病毒载量34例,采用提取柱法与磁珠法核酸定量检测,根据浓度1×10^(8)样本梯度稀释结果比较2种核酸定量检测方法的灵敏度和线性范围.[结果]磁珠法HCV核酸定量检测阳性率为70.0%,与提取柱法的66.7%比较差异无统计学意义(P>0.05);22例低病毒载量的HCV血清标本磁珠法检测阳性率为40.9%,明显高于提取柱法的9.1%(χ^(2)=5.9394,P=0.0148);磁珠法灵敏度和线性范围分别为39 U/mL和3.9×(10^(1)~10^(8)),与提取柱法比较差异有统计学意义(P<0.05).[结论]磁珠法与提取柱法在中、高病毒载量的检测中一致性较好,在低病毒载量的检测中磁珠法较提取柱法核酸定量检测的灵敏度更高,线性范围更广.OBJECTIVE To observe the application effect of magnetic bead method in the nucleic acid quantitative detection of hepatitis C virus.METHODS Serum specimens of hepatitis C virus(HCV)from 120 patients with hepatitis C were collected from January 2018 to January 2020,including 22 cases with low viral load,64 cases with medium viral load and 34 cases with high viral load.The extraction column and magnetic bead method were used to detect nucleic acid quantitatively.The sensitivity and linear range of the two methods were compared according to the result of 1×10^(8) sample concentration gradient dilution.RESULTS The positive rate of HCV nucleic acid quantitative detection by magnetic bead method was 70.0%,66.7%by extraction column method,the difference was not statistically significant(P>0.05).The positive rate of 22 HCV serum samples with low viral load was 40.9%by magnetic bead method,which was significantly higher than 9.1%by extraction column method(χ^(2)=5.9394,P=0.0148).The sensitivity and linear range of the magnetic bead method were 39 U/mL and 3.9×(10^(1)-10^(8))respectively,which were significantly different from the extraction column method(P<0.05).CONCLUSION The magnetic bead method and the extraction column method have good consistency in the detection of medium and high viral load,while magnetic bead method has higher sensitivity and wider linear range than extraction column method in the detection of low viral load.
