补肾助孕方对肾虚薄型子宫内膜大鼠组织形态ERa、PR表达影响及容受性的调控作用

摘　　要：目的观察补肾助孕方对肾虚薄型大鼠子宫内膜、卵巢组织形态影响及ERa、PR表达变化,探讨中药对子宫内膜容受性的调控作用。方法雌性SD大鼠30只随机分为正常组、模型组、中药组,每组10只。后两组经羟基脲灌胃建立肾虚薄型子宫内膜大鼠模型。模型成功后,中药组给予补肾助孕方0.3 mg/(kg·d)灌胃,对照组和模型组给予等体积生理盐水,持续15 d。干预结束后,HE染色观察大鼠子宫内膜及卵巢组织形态;Elisa检测血液中孕酮及雌二醇水平;免疫组化及Western blot法检测大鼠子宫内膜ERa、PR表达。结果与正常组比较,模型组大鼠子宫内膜腺体稀少,分泌功能降低,内膜间质致密,卵巢组织内卵泡数目减少,卵泡内细胞层数减少,颗粒细胞排列无序;孕酮及雌二醇水平减低,ERa、PR表达水平降低,差异有统计学意义(P<0.01)。与模型组比较,中药组子宫内膜腺体数目增多,分泌增加,间质蜕膜化,卵巢内卵泡数目增加,细胞层数增加,颗粒细胞排列较规则;孕酮及雌二醇水平升高,ERa、PR表达水平升高,差异有统计学意义(P<0.05)。结论补肾助孕方素能改善肾虚薄型大鼠子宫内膜及卵巢组织形态,并上调雌、孕激素的表达,调控种植窗促进胚胎植入。Objective To observe the effects of Bushen Zhuyun Recipe on the morphology of endometrial and ovarian tissues and the expression of ERa and PR in rats with kidney deficiency and thin type,and to explore the regulatory effect of traditional Chinese medicine on endometrial receptivity.Methods Thirty female SD rats were randomly divided into normal group,model group and traditional Chinese medicine group,with 10 rats in each group.The latter two groups were given hydroxyurea gavage to establish a rat model of kidney deficiency and thin endometrium.After the model was successful,the Chinese medicine group was given the Bushen Zhuyun prescription 0.3 mg/kg/d by gavage,and the control group and the model group were given equal volume of normal saline for 15 days.After the intervention,HE staining was used to observe the morphology of rat endometrial and ovarian tissue;Elisa was used to detect the levels of progesterone and estradiol in the blood;immunohistochemistry and WB methods were used to detect the expression of ERa and PR in rat endometrial.Results Compared with the control group,the model group had fewer endometrial glands,decreased secretory function,dense endometrial stroma,decreased number of follicles in the ovarian tissue,cystic expansion,and thinner granular cell layers;progesterone and estradiol The alcohol level decreased,and the expression of ERa and PR decreased,and the difference was statistically significant(P<0.01).Compared with the model group,the number of endometrial glands in the traditional Chinese medicine group increased,secretion increased,interstitial decidualization,the number of follicles in the ovary increased,the number of cell layers increased;the levels of progesterone and estradiol increased,and the expression of ERa and PR Increase,the difference was statistically significant(P<0.05).Conclusion Bushen Zhuyun decoction can improve the endometrial and ovarian tissue morphology of rats with kidney deficiency,upregulate the expression of ER and PR,and regulate the endometrial receptivi
