摘  要:目的探讨肉苁蓉苯乙醇总苷(CPhGs)对腹主动脉缩窄术(AAC)后心肌肥厚大鼠的作用及其作用机制。方法将70只实验大鼠分为7组:正常对照组、模型组、假手术组、缬沙坦阳性药物组(8.3 mg/kg)、模型+CPhGs低剂量组(125 mg/kg)、模型+CPhGs中剂量组(250 mg/kg)、模型+CPhGs高剂量组(500 mg/kg),各组10只。对模型组大鼠行AAC术,使大鼠腹主动脉管腔环形缩窄,构建心肌肥厚模型,术后每天灌胃给药1次,持续6周。各组以超声心动图检测大鼠心率(HR)、收缩期间隔厚度(IVSs)、左室收缩末期内径(LVESD)左室厚壁舒张期厚度(LVPWd)、射血分数(EF),碱水解法测量羟脯氨酸(HYP),苏木精-伊红(HE)染色观察心肌细胞的表型、数目、排列方式并进行比较。结果与正常对照组相比,模型组大鼠HR、IVSs、LVPWd、HYP均有不同程度增加(P均<0.05),LVESD均有不同程度的降低(P均<0.05)。与模型组相比,模型+CPhGs低、中、高组大鼠HR、IVSs、LVPWd、HYP均有不同程度下降(P均<0.05),LVESD、EF均有不同程度的升高(P均<0.05),模型+CPhGs高剂量组与缬沙坦阳性药物组各项指标差异均无统计学意义。结论CPhGs可以抑制AAC术后心肌肥厚的发生,起到保护心肌的作用。Objective To observe the protective effect of total glycosides of Cistanche phenylethanoid glycosides(CPhGs)on myocardial hypertrophy rats after abdominal aorta constriction(AAC).Methods 70 rats were divided into 7 groups:normal control group,model group,sham operation group,positive drug control group(valsartan 8.3 mg/kg),model+CPhGs(125 mg/kg,250 mg/kg,500 mg/kg).The AAC method was used to cause coarctation of the abdominal aortic lumen,which led to the formation of hypertensive myocardial hypertrophy model in rats.After the operation,the rats were administrated by lavage once a day for 6 weeks.Blood pressure(BP)was measured by noninvasive sphygmomanometer;The changes of HR,IVSS,LVESD,LVPwd and EF were measured by echocardiography;The content of hydroxyproline(HYP)in myocardial homogenate was determined;The pathological changes of myocardial cells were observed after HE staining.Results Compared with the normal control group,HR,IVSs,LVPWd and HYP of the model group were increased in varying degrees(P<0.05);LVESD,EF,SBP and DBP were decreased in varying degrees(P<0.05);Compared with the model group,HR,IVSs,LVPWd and HYP of the model+CPhGs low,medium and high groups were decreased in varying degrees(P<0.05),and LVESD,EF,SBP and DBP of the the model+CPhGs low,medium and high groups were increased in varying degrees(P<0.05).But there was no significant difference between the model+CPhGs high group and valsartan positive drug group.Conclusion CPhGs can inhibit the inducing factors of cardiac hypertrophy induced by AAC,improve cardiac function and protect myocardium.
