摘　　要：目的:研究载他克莫司可注射温敏水凝胶(FK506-TH)对人牙髓干细胞(hDPSCs)成骨分化的影响。方法:酶消化法体外分离培养hDPSCs,CCK-8法检测不同浓度FK506FK506-TH对hDPSCs增殖的影响,采用Real-time PCR、茜素红S和碱性磷酸酶(ALP)染色检测各组细胞成骨分化能力的影响。结果:FK506-TH能促进hDPSCs增殖(P<0.05),Real-time PCR数据表明,FK506-TH组细胞成骨基因的mRNA均明显高于FK506药物组和单纯水凝胶组(P<0.05),且FK506-TH细胞矿化结节及ALP活性较FK506药物组和单纯水凝胶组明显增加(P<0.001)。结论:FK506-TH可提高hDPSCs的增殖能力以及成骨分化的潜能。Objective:To study the effects of injectable temperature sensitive FK506 hydrogel(FK506-TH)on the osteogenic differentiation of human dental pulp stem cells(hDPSCs).Methods:hDPSCs were in vitro cultured,the effects of FK506-TH on hDPSCs proliferation was detected by CCK-8 assay,the osteogenic differentiation ability of the cells was detected by RT-PCR,alizarin red staining and alkaline phosphatase(ALP)staining.Results:FK506-TH promoted the proliferation of hDPSCs(P<0.05).RT-PCR data showed that the mRNA expression of osteogenesis related genes was significantly higher in FK506-TH group than that of FK506 and hydrogel alone groups(P<0.05),the mineralization and ALP were significantly higher than those of FK506 or hydrogel alone groups(P<0.001).Conclusion:FK506-TH can promote the proliferation and osteogenic differentiation potential of hDPSCs.
