摘 要:目的应用网络药理学方法探究桃红四物汤治疗类风湿性关节炎潜在药效作用机制。方法基于课题组前期UPLC-Q-TOF-MS E数据并结合TCMSP数据库,获取桃红四物汤主要化学成分,将口服生物利用度(OB)、类药性(DL)设定为筛选参数,寻找活性成分,通过PharmMapper数据库进行靶点预测;从OMIM、GeneCards数据库对类风湿性关节炎的靶点进行检索;使用Cytoscape 3.7.1软件构建成分-靶点相互作用网络,对该网络进行拓扑学分析得到核心活性成分;使用STRING数据库绘制蛋白-蛋白相互作用网络,对该网络进行拓扑学分析得到核心靶点;对交集靶点基因利用DAVID数据库进行GO分析和KEGG分析,探讨潜在靶点可能具有的生物学功能及通路。结果从桃红四物汤中筛选出38个活性成分,核心成分11个;治疗类风湿性关节炎的靶点291个,核心靶点33个;GO分析结果显示59个生物过程、32个分子功能、15个细胞组成;KEGG分析结果显示34条信号通路。结论本研究初步证实桃红四物汤通过多靶点、多通路治疗类风湿性关节炎,为类风湿性关节炎的治疗提供思路和参考。Aim To explore the possible mechanism of Tao-Hong-Si-Wu decoction in the treatment of rheumatoid arthritis using network pharmacology.Methods Based on the previous UPLC-Q-TOF-MS E data in combination with TCMSP,the main chemical compositions in Tao-Hong-Si-Wu decoction were obtained.Oral absorption rate(OB)and drug-like property(DL)were set as screening parameters to find the active components.PharmMapper database was used to find potential targets;the target of rheumatoid arthritis was retrieved from OMIM,GeneCards databases;the interaction network of ingredient-target was constructed by Cytoscape 3.7.1 software,and the core chemical constituents were searched by network topology;STRING database was used to draw protein-protein interaction network;DAVID database was used for GO analysis and KEGG analysis to explore the biological functions and pathways of potential targets.Results Altogether 38 active components were screened from Tao-Hong-Si-Wu decoction,including 11 core components;291 targets for rheumatoid arthritis treatment,including 33 core targets;GO analysis showed that they involved 59 biological processes,11 molecular functions and 13 cellular components;KEGG analysis showed that they involved 34 signaling pathways.Conclusions Tao-Hong-Si-Wu decoction can treat rheumatoid arthritis through multi-targets and multi-pathways,which provides ideas and references for the future research on the treatment of rheumatoid arthritis.
