摘  要:为研究鳜Siniperca chuatsi视觉转变期视网膜感光细胞层结构变化,利用组织切片、免疫荧光技术与几何构型分析对鳜视觉转变期3个阶段(出膜后第28、42、63天,记为D_(28)、D_(42)和D_(63))视网膜中感光细胞数量增长方式和视锥细胞排布进行了研究。结果表明:鳜视觉转变期视网膜各区域视杆细胞数量显著增加,D_(63)时期已达4500~6000 cells/100μm^(2);单锥细胞(SC)数量上升缓慢,D_(28)时期为20~40 cells/100μm^(2),而D_(63)时为5~8 cells/100μm^(2),同时双锥细胞(DC)数量增长速度明显快于单锥细胞,D_(28)时期为70~100 cells/100μm^(2),而D_(63)时期为100~150 cells/100μm^(2),视锥细胞中双锥细胞比例显著上升,D_(63)时期DC/SC已接近50;视锥细胞在视网膜腹侧的增长均快于背侧,而增长的视杆细胞未影响视锥细胞排布的构建。研究表明,鳜视觉转变期视网膜感光细胞层在结构上发生了剧烈变化,视杆细胞在视网膜各区域均匀增长,视锥细胞腹侧增长速度明显快于背侧,鳜感光细胞发育特点为其底栖暗光生活的视觉转变提供了结构基础。Growth pattern and cone mosaic in retina were investigated in mandarinfish Siniperca chuatsi during three stages of visual transition period(D_(28)(28 days post-hatching),body length of 25-35 mm,D_(42)(42 days post-hatching),and D_(63)(63 days post-hatching),body length of 85-100 mm)by means of histological method,immunofluorescence and geometry analysis in order to evaluate the retina photoreceptor layer structural changes in mandarinfish during the visual transition period.It was found that there was significant increase in number of rod cells during the visual transition period,4500-6000 cells/100μm^(2) in each area in D_(63);slow increase in the single cone cell(SC)number,20-40 cells/100μm^(2) in D28,only 5-8 cells/100μm^(2) in D_(63).Meanwhile,there was more rapid growth of double cone cell(DC)than that of SC,with 70-100 cells/100μm^(2) in D28,and 100-150 cells/100μm^(2) in D_(63),as a whole the DC/SC ratio was decreased,close to 50 in D_(63).There was better growth of cone cells in the ventral side than that in the dorsal side,and the rod cell rapid growth did not affect cone mosaic formation.The structure of retina photoreceptor layer was changed dramatically during the visual transition period,with significant change in number and proportion of all kinds of photoreceptor cells.There were evenly development of rod cells and regionalization growth of cone cells,with two development events independent of each other which together to provide a structural basis for its visual transformation.The findings indicated that the photoreceptor layer of mandarinfish retina was changed dramatically in the visual transition period,the rod cells grew evenly in all regions,and the cone cells on the ventral side grew faster than that on the dorsal side.These developmental characteristics of photoreceptor cells provide the structural basis for the visual transformation of mandarinfish in benthic living habit under dim environment.
