摘 要:为了对巢湖蓝藻进行更高效地资源化利用,文章以羟基磷灰石为金属螯合亲和色谱填料,以透析脱盐后的藻蓝蛋白初步纯化溶液为样品,考察了洗脱液pH值、离子强度、洗脱速度对藻蓝蛋白纯化的影响。通过响应面确定了最佳工艺参数为洗脱液pH值6.32、离子强度0.08 mol/L、洗脱速度3.56 mL/min。同时对藻蓝蛋白进行紫外-可见光谱、荧光光谱和红外光谱分析,表明所选工艺适合提纯试剂级藻蓝蛋白。在此条件下,藻蓝蛋白纯度与回收率的理论最佳值为4.52%和38.36%,3次实验均值为4.51%、38.38%,纯化效果优于单因素实验结果。In order to make more efficient resource utilization of Chaohu cyanobacteria,this research,taking hydroxyapatite as metal chelate affinity chromatographic packing,used preliminarily purified phycocyanin solution after dialysis desalting as the sample,examined the effect of the pH,ionic strength,elution velocity of eluent on phycocyanin purification.According to response surface methodology(RSM),the optimum technological parameters were determined as follows:the eluent pH was 6.32,the ionic strength was 0.08 mol/L,and the elution velocity was 3.56 mL/min.At the same time,the phycocyanin was analyzed by UV-VIS,Fluorescence and infrared spectra,which showed that the selected technology was suitable for purification of reagent grade phycocyanin.Under these conditions,the theoretical optimal values of purity and recovery of phycocyanin were 4.52%and 38.36%,respectively,and the mean values of three repeated experiments were 4.51%and 38.38%.The purification effect is better than that of single factor experiment.
