摘 要:目的:基于原子力显微技术,建立子宫内膜异位症细胞力学性质检测方法,比较子宫内膜异位症在位与异位子宫内膜细胞的微尺度刚度,以进一步研究子宫内膜异位症发病机制。方法:采集子宫内膜异位症患者子宫内膜及其异位到卵巢的巧克力囊肿囊皮,并分别进行原代培养,利用原子力显微镜采集细胞力曲线,获取杨氏模量信息,进行统计分析。结果:在位腺上皮细胞的杨氏模量高于异位腺上皮细胞(2.96±1.48 kPa,1.47±1.15 kPa,P<0.05);在位间质细胞与异位间质细胞杨氏模量比较无明显变化(3.93±1.50kPa,3.14±1.65kPa,P>0.05)。结论:子宫内膜在位腺上皮细胞与异位腺上皮细胞力学特性不同,异位腺上皮细胞刚度降低,因此推论细胞刚度小更有利于细胞的迁移,本研究结果支持细胞迁移学说。Objective: The mechanical model of endometrial cell was established based on atomic force microscopy(AFM), and the microscale stiffness of endometriosis was compared with endometrial cells in order to further study the pathogenesis of endometriosis. Methods: Endometrial tissue and the wall of a chocolate cyst of EMs were acquired, the primary culture of endometriosis tissue was carried out, AFM was used to obtain the cell force curve, and the apparent Young’s modulus of the cell calculated. Results: Young’s modulus of locational glandular epithelial cells is higher than that of the ectopic glandular epithelial cells [(2.96±1.48)kPa,(1.47±1.15)kPa, P<0.05];Young’s modulus of stromal cells of normal position is compared with Young’s modulus of stromal cells of abnormal position, and there are no obvious change [(3.93±1.50)kPa,(3.14±1.65)kPa, P>0.05]. Conclusion: These findings suggest that the mechanical properties are different between the two kinds of cells, the stiffness of ectopic epithelial cells is much smaller, and the small cell stiffness is more conducive to cell migration. Therefore, this study also supports the cell migration theory.
