摘　　要：为了研究豫南黑猪仔猪近年来发生毛色变异比例逐年升高的遗传因素,试验选择8头具有4种毛色的豫南黑猪及其4头母本(黑色毛色)和14头黑色毛色的豫南黑猪作为样本,采用PCR扩增和测序的方法对MC1R基因的全长序列进行分析,并与欧亚野猪等具有代表性毛色猪种的MC1R基因序列比较。结果表明:26头豫南黑猪MC1R基因全长序列中共鉴定到13个编码区突变和7个非翻译区(untranslated region,UTR)突变。其中2个编码区突变为新发现的突变,分别位于第84个密码子(GCC)的g.252 C>T和第138个密码子(CTG)的g.414 G>A,均为同义突变。豫南黑猪的MC1R基因存在欧亚野猪、欧洲商品猪及几个中国地方猪已知的毛色相关突变位点。全红和全黄豫南黑猪个体具有杜洛克猪的特有突变,黄底黑斑和红底黑斑豫南黑猪个体中存在皮特兰猪中的特有突变,初步判定毛色异常的个体母本渗入了外来猪种的血统。说明MC1R基因突变是导致豫南黑猪毛色出现异常的遗传原因。In order to study the genetic factors of increasing hair color variation ratio year by year in the piglets of Yunan black pigs in recent years,eight Yunan black pigs with four kinds of abnormal hair color,their four mothers with black hair color and 14 individuals with black hair color were selected as the test samples,then the full-length sequence of MC1 R gene were analyzed by PCR amplification and sequencing method and compared with the MC1 R gene sequence of Eurasian wild boar with representative coat colors.The results showed that 13 coding region mutations and 7 untranslated region(UTR)mutations were identified from the full-length sequence of the MC1 R gene in 26 Yunan black pigs.Among them,the mutations in 2 coding regions were newly discovered,located at g.252 C>T of the 84 th codon(GCC)and g.414 G>A of the 138 th codon(CTG),respectively,which were synonymous mutations.The MC1 R gene of Yunan black pigs contained known mutations related to hair color in Eurasian wild boars,European commercial pigs and several Chinese native pigs.Yunan black pigs with all yellow and all red hair color had the unique mutation of Duroc,and Yunan black pigs with black spots on red or yellow background had the unique mutation of Pietrain.It was preliminarily determined that the lineage of foreign pig breeds infiltrated into the female parent with abnormal coat color.The results indicated that mutatien of MC1 R gene was the genetic cause of abnormal coat color of Yunan black pigs.
