摘 要:为了研究枫蓼提取物中黄酮类作为止泻的药效物质基础,试验采用超高效液相-飞行时间质谱(UHPLC-Q-TOF-MS)分析方法,对枫蓼提取物的化学成分进行鉴定;利用BL-420S生物机能系统,以家兔离体肠平滑肌收缩张力和频率为指标,观察提取物及单体成分对肠平滑肌收缩张力及收缩频率的影响。结果表明:枫蓼水提物和醇提物中共鉴定了44种化合物,黄酮类、多酚类是主要成分之一,且醇提物的峰面积积分高于水提物。离体肠平滑肌收缩药效学表明,与给药前比,给药后水提物、醇提物中剂量(0.25 g/mL)、高剂量(0.5 g/mL)使空肠自主收缩张力显著或极显著下降(P<0.05或P<0.01),水提物高剂量使空肠收缩频率显著增加(P<0.05);高剂量水提物和醇提物均可显著降低回肠收缩张力(P<0.05),各剂量水提物和醇提物对回肠收缩频率均无显著影响(P>0.05)。在乙酰胆碱引起的肠平滑肌痉挛模型中,与给药前比,给药后各剂量水提物、醇提物均可使空肠平滑肌收缩张力极显著下降(P<0.01),醇提物对收缩频率的影响呈剂量依赖性;高剂量水提物使回肠平滑肌收缩张力显著下降(P<0.05),各剂量醇提物均可使回肠平滑肌收缩张力极显著下降(P<0.01),给药前后回肠收缩频率均无显著影响(P>0.05)。在此基础上,对其中的槲皮素、芦丁、槲皮苷等成分进行相应的药效学研究,槲皮素、芦丁、山柰酚、异鼠李素200,400μg,槲皮苷200μg、金丝桃苷400μg均可抑制由乙酰胆碱引起的空肠平滑肌痉挛,与给药前比,给药后收缩张力显著下降(P<0.05);槲皮苷400μg、金丝桃苷200μg对收缩张力无显著影响(P>0.05);槲皮素、山柰酚200,400μg均可抑制由乙酰胆碱引起的回肠平滑肌痉挛,与给药前比,给药后收缩张力显著下降(P<0.05),芦丁、槲皮苷、金丝桃苷、异鼠李素200,400μg对收缩张力均没有显著影响(P>0.05)。各处理对空肠和回肠的收缩频率均无�In order to study the flavonoids in the Fengliao extracts as a basis of anti-diarrheal drug,ultra performance liquid chromatography-time-of-flight mass spectrometry(UHPLC-Q-TOF-MS)was used in this experiment to identify the chemical components of the Fengliao extracts.Using the BL-420 S biological function system,the contraction tension and frequency of rabbit intestinal smooth muscle were used as indexes to observe the effect of extracts and monomer components on the contraction tension and frequency of intestinal smooth muscle.The results showed that a total of 44 compounds were identified in the Fengliao water extracts and alcohol extracts.Flavonoids and polyphenols were the main components,and the peak area integral of the alcohol extracts was higher than that of the water extracts.The pharmacodynamics results of isolated intestinal smooth muscle contraction demonstrated that compared with before administration,the middle(0.25 g/mL)and high(0.5 g/mL)doses of water and alcohol extracts significantly(P<0.05)or extremely significantly(P<0.01)decreased the jejunal spontaneous contraction tension after administration.The high dose of water extracts significantly(P<0.05)increased the jejunal contraction frequency.High-dose water extracts and alcohol extracts could significantly reduce ileal contraction tension(P<0.05),and there was no significant effect on the frequency of ileal contraction(P>0.05)among each dose of water extracts and alcohol extracts.In the model of acetylcholine-induced intestinal smooth muscle contraction,compared with before administration,each dose of the water and alcohol extracts can very significantly(P<0.01)reduce contraction tension of jejunal smooth muscle after administration.The alcohol extracts had a dose-dependent effect on the contraction frequency.High-dose water extracts significantly reduced the contraction tension of ileal smooth muscle(P<0.05),and each dose of alcohol extracts could significantly reduce the contractile tension of ileal smooth muscle(P<0.01).There was no signifi
