摘 要:目的研究多巴胺D1受体拮抗剂SCH23390对吗啡戒断反应和突触素(SYN)的影响。方法将40只大鼠按随机数字表法分为对照组、吗啡组、SCH23390组、SKF38393组,每组10只。吗啡组、SCH23390组和SKF38393组大鼠按剂量递增方式注射吗啡5 d建立吗啡暴露动物模型。SCH23390组和SKF38393组大鼠在第1~4天上午腹腔注射吗啡前15 min中脑导水管灰质区注射0.5μL 10 mmol/L SCH23390或0.5μL 5 mmol/L SKF38393,在第5天所有大鼠注射纳络酮观察戒断反应,免疫组化和免疫印迹方法检测SYN水平。结果吗啡组戒断反应总分及齿颤、腹泻、反常姿势、眯眼、湿狗样抖动评分高于对照组,差异有统计学意义(P<0.05或P<0.01)。SCH23390组戒断反应总分及齿颤、腹泻评分低于吗啡组,差异有高度统计学意义(P<0.01)。SKF38393组和吗啡组戒断反应总分及齿颤、腹泻、眯眼、湿狗样抖动评分比较,差异无统计意义(P>0.05);而SKF38393组反常姿势评分高于吗啡组,差异有统计学意义(P<0.05)。吗啡组SYN蛋白表达水平高于对照组,差异有高度统计学意义(P<0.01)。SCH23390组SYN蛋白表达水平低于吗啡组,差异有统计学意义(P<0.05)。SKF38393组SYN蛋白表达高于吗啡组,差异有统计学意义(P<0.05)。吗啡组SYN阳性神经元数量高于对照组,差异有高度统计学意义(P<0.01)。SCH23390组SYN阳性神经元数量低于吗啡组,差异有统计学意义(P<0.05)。SKF38393组SYN阳性神经元数量高于吗啡组,差异有统计学意义(P<0.05)。结论SCH23390可能通过下调SYN减轻吗啡戒断反应。Objective To study the effect of dopamine D1 receptor antagonist SCH23390 on Morphine withdrawal response and synaptophysin(SYN)expression.Methods According to the random number table method,40 rats were divided into control group,Morphine group,SCH23390 group and SKF38393 group,with 10 rats in each group.Rats in Morphine group,SCH23390 group and SKF38393 group were injected with Morphine in increasing doses for 5 days to establish Morphine-exposure animal models.Rats in SCH23390 group and SKF38393 group were injected with 0.5μL 10 mmol/L SCH23390 or 0.5μL 5 mmol/L SKF38393 into the midbrain periaqueductal gray 15 min before intraperitoneal Morphine injections on the mornings of days 1-4.On the 5th day,all rats were injected with Naloxone to observe withdrawal response,and SYN level was detected by immunohistochemistry and Western blotting.Results Withdrawal response total score and scores of tooth tremor,diarrhea,abnormal posture,squinting and wet dog shaking in Morphine group were higher than those in control group,and the differences were statistically significant(P<0.05 or P<0.01).Withdrawal response total scores and scores of tooth tremor and diarrhea in SCH23390 group were lower than those in Morphine group,and the differences were highly statistically significant(P<0.01).There was no significant difference in withdrawal response total scores and scores of tooth tremor,diarrhea,squinting and wet dog shaking between SKF38393 group and Morphine group(P>0.05);while score of abnormal posture of SKF38393 group was higher than that of Morphine group,and the difference was statistically significant(P<0.05).The expression level of SYN protein in Morphine group was higher than that in control group,and the difference was highly statistically significant(P<0.01).The expression level of SYN protein in SCH23390 group was lower than that in Morphine group,and the difference was statistically significant(P<0.05).The expression level of SYN protein in SKF38393 group was higher than that in Morphine group,and the differe
