电针通过抑制氧化应激和炎症反应减轻DSS诱导的大鼠结肠炎

摘　　要：目的探讨电针对葡聚糖硫酸钠(dextran sodium sulfate,DSS)诱导的大鼠结肠炎的治疗作用及机制。方法大鼠口服DSS诱发慢性结肠炎,接受为期21d的电针治疗后,剪取结肠并测量长度,同时检测血清中LDH的水平和结肠屏障功能;HE染色评估结肠组织的病理学改变情况;TUNEL染色和cleaved caspase-3免疫组织化学染色评估结肠组织损伤情况;试剂盒法检测结肠组织氧化应激指标MDA和GSH水平以及SOD和iNOS活性;ELISA法检测结肠组织中促炎因子IL-1β、IL-6和TNF-α水平;Western blot法检测结肠组织中claudin-3和ZO-1水平。结果21d的电针治疗明显改善DSS引起的结肠缩短、结肠屏障破坏、血清LDH释放和病理学损伤。电针治疗能有效恢复DSS处理的大鼠结肠组织中claudin-3和ZO-1水平,以及降低结肠组织中TNF-α、IL-1β、IL-6水平,并抑制iNOS和MDA活性,同时增加GSH和SOD的活性。结论电针治疗可通过修复肠屏障功能、降低炎症反应和氧化应激来减轻DSS诱发的大鼠慢性结肠炎。Objective To investigate the therapeutic effects and mechanisms of electroacupuncture on DSS(dextran sulfate sodium)-induced colitis in rats.Methods Chronic colitis was induced by oral DSS administration to rats,followed by electroacupuncture treatment for 21 days.At the end of treatment,the colon was clipped and the length was measured.Meanwhile,serum LDH level and colon barrier function were detected.The pathological changes in colon tissues were evaluated by HE staining.Colonic tissue damages were assessed by TUNEL staining and cleaved Caspase-3 immunohistochemical staining.The indexes of oxidative stress including MDA and GSH levels as well as SOD and iNOS activities in colon tissues were measured by the Reagent Kit method.The levels of pro-inflammatory factors IL-1β,IL-6 and TNF-αin colon tissues were determined by ELISA.The expression levels of claudin-3 and ZO-1 proteins in colon tissues were detected by Western blot.Results The 21-day electroacupuncture procedure attenuated DSS-induced colon shortening,colon barrier destruction,serum LDH release,and pathological injury.Electroacupuncture treatment effectively restored the expressions of claudin-3 and ZO-1,reduced the levels of IL-1β,IL-6 and TNF-α,inhibited the levels of iNOS and MDA,and increased the activities of GSH and SOD in the colon tissues of DSS-treated rats.Conclusion Electroacupuncture therapy can alleviate chronic colitis induced by DSS in rats by repairing colon barrier function,reducing inflammatory response and oxidative stress.
