摘  要:目的探讨10-姜酚对大鼠缺血性心律失常及其心肌组织miRNA-29表达的影响。方法SPF级6周龄SD大鼠,按照体重随机分为对照组、模型组、阳性给药组(盐酸普萘洛尔2 mg·kg^(-1))、10-姜酚给药组(8、4、2 mg·kg^(-1))。除对照组之外,其余各组大鼠均尾静脉注射5μg·kg^(-1)的乌头碱生理盐水溶液,诱发大鼠缺血性心律失常模型。随后阳性给药组和10-姜酚组分别尾静脉给药,对照组和模型组尾静脉注射生理盐水,给药30 min内记录各组大鼠心电图变化,早搏早搏(VPB)次数以及首次出现时间,室速(VT)和室颤(VF)持续次数和时间。待实验结束后取大鼠心脏,测定心肌梗死面积,计算心肌梗死率。qRT-PCR检测各组大鼠心肌组织miRNA-29的表达。结果与对照组相比,乌头碱诱导模型组出现明显的缺血性心律失常,VPB、VT和VF频率明显增加(P<0.05),VPB首次出现时间显著缩短(P<0.05),VT和VF持续时间也显著增长(P<0.05),模型组大鼠心肌组织梗死面积明显增多(P<0.05),心肌组织miRNA-29表达降低(P<0.05);给药治疗后,与模型组相比,10-姜酚可以显著改善大鼠出现的缺血性心律失常,VPB、VT和VF频率明显降低(P<0.05);VPB首次出现时间推迟(P<0.05),VT和VF持续时间也显著降低(P<0.05),心肌组织梗死面积明显减少(P<0.05),心肌组织miRNA-29表达降低(P<0.05)。结论10-姜酚可以显著改善由乌头碱诱导大鼠缺血性心律失常,同时显著降低心肌组织中miRNA-29的表达水平。OBJECTIVE To investigate the effect of 10-gingerol on ischemic arrhythmia and expression of miRNA-29 in myocardial cells of rats.METHODS SPF 6-week-old SD rats were randomly divided into control group,model group and positive drug group(propranolol hydrochloride 2 mL·kg^(-1)),10-gingerol administration group(8,4,2 mg·kg^(-1)).In addition to the control group,rats in the other groups were injected with 5μg·kg^(-1) of aconitine physiological saline solution in the tail vein to induce a rat model of ischemic arrhythmia.Subsequently,the positive drug-administered group and the 10-gingerol group were administered respectively via tail vein,and the control group and the model group were injected with physiological saline via tail vein.The electrocardiogram changes,the number of premature beats(VPB)and the first appearance time of each group were recorded within 30 minutes after administration,ventricular velocities(VT)and ventricular fibrillation(VF)duration and time.At the end of the experiment,the rat heart was taken,the myocardial infarct size was measured,and the expression of tissue miRNA-29 was detected by qRT-PCR.RESULTS Compared with the control group,the aconitine-induced model group showed obvious ischemic arrhythmia,the frequencies of VPB,VT and VF increased significantly(P<0.05),and the first time of VPB appeared significantly shortened(P<0.05).The duration of VF and VF also increased significantly(P<0.05).The infarct size of myocardial tissue in the model group was significantly increased(P<0.05),and the expression of miRNA-29 in myocardial tissue was decreased(P<0.05).After treatment,the model group in comparison,10-gingerol significantly improved the ischemic arrhythmia in rats,and the frequencies of VPB,VT and VF were significantly decreased(P<0.05);the first time of VPB was delayed(P<0.05),and the duration of VT and VF was also decreased(P<0.05),myocardial infarct size was significantly reduced(P<0.05),and myocardial tissue miRNA-29 expression was decreased(P<0.05).CONCLUSION 10-gingerol can signif
