强化血糖、血压控制对糖尿病足患者外泌体效应microRNA的影响

摘　　要：目的观察糖尿病患者治疗前后血外泌体分泌及效应因子miR-126-3P、miR-125b-1-3p水平变化,探讨强化血糖、血压控制对糖尿病足溃疡创面愈合的作用机制。方法30例糖尿病足患者分别于强化治疗1周前后采血,抽提外泌体,行外泌体鉴定,观察外泌体分泌情况并测定miR-126-3P、miR-125b-1-3p水平。结果强化血糖、血压治疗后,糖尿病足患者血外泌体miR-126-3P、miR-125b-1-3p水平升高(P<0.05);相关性分析显示miR-126-3P水平与空腹血糖(FBG)呈正相关关系(r=0.37,P<0.05)。结论强化血糖、血压控制可升高糖尿病足患者外泌体miR-126-3P、miR-125b-1-3p水平,可能对糖尿病足溃疡创面愈合具有积极意义。Objective To observe serum exosomes secretion and the changes of levels of mir-126-3p and mir-125b-1-3p in diabetic patients before and after treatment,and to explore the mechanism of intensive blood glucose and blood pressure control on the wound healing of the diabetic foot ulcers.Methods Blood collection,exosomes extraction and identification were respectively performed on 30 patients with diabetic foot before and after intensive treatment for 1 week.Exosomes were extracted and identified.And then,the secretion of exosomes was observed and the levels of miR-126-3p and miR-125b-1-3p were measured.Results After intensive blood glucose and blood pressure treatment,blood exosomes miR-126-3P and miR-125b-1-3p levels in diabetic foot patients increased(P<0.05);and correlation analysis showed that miR-126-3P levels were positively correlated with fasting blood glucose(FBG)(r=0.37,P<0.05).Conclusion Intensive blood sugar and blood pressure control can increase the levels of exosomes miR-126-3P and miR-125b-1-3p in diabetic foot patients,which may have positive significance for the wound healing of diabetic foot ulcers.
