摘 要:目的通过研究黄芩、大黄、黄连三种中药不同复方配伍对脂多糖(lipopolysaccharide,LPS)刺激下人牙周膜细胞前列腺素E_(2)(prostaglandin E_(2),PGE_(2))的影响,初步筛选出具有增效减毒治疗牙周炎的复方配伍。方法采用超声萃取技术分别获得黄芩、大黄、黄连有效部位提取物,应用高效液相色谱(high performance liquid chromatography,HPLC)技术测定其有效成分含量;三种中药各自选择不同浓度,分别为黄芩0,0.5,5,50 mg/L,大黄0,0.13,1.3,13 mg/L,黄连0,0.012,0.12,1.2 mg/L,按照正交实验法进行配伍,采用酶联免疫吸附实验(enzyme-linked immunosorbent assay,ELISA)分别检测不同药物配伍对LPS刺激下人牙周膜细胞(human periodontal ligament cells,HPDLCs)产生PGE 2的影响。结果经HPLC分析,中药黄芩、大黄和黄连的各有效部位中黄芩素、大黄素、盐酸小檗碱的含量依次为19.94%,7.50%,80.87%;交叉配伍的32组复方溶液中得出5组配伍对LPS刺激下HPDLCs的PGE_(2)表达有明显抑制作用,分别是5 mg/L黄芩乙醚部位和0.12 mg/L黄连水部位配伍,13 mg/L大黄乙醚部位和0.012 mg/L黄连水部位配伍,13 mg/L大黄乙醚部位和0.50 mg/L黄芩乙醚部位配伍,13 mg/L大黄乙醚部位和5 mg/L黄芩乙醚部位配伍,50 mg/L黄芩乙醚部位。结论黄芩、大黄、黄连三种中药复方配伍后有5组配伍能抑制LPS刺激HPDLCs产生PGE_(2)的含量,为后续体内实验剂量的选用提供依据。Objective To study the effect of different combinations of Scutellaria,Rhubarb and Coptis on prostaglandin E_(2)(PGE_(2))produced by lipopolysaccharide(LPS)-stimulated human periodontal ligament cells(HPDLCs),and explore the effective combinations in the treatment of periodontitis.Methods The effective components of Scutellaria,Rhubarb and Coptidis were extracted by ultraso-nic extraction,and the contents of the effective components were determined by high performance liquid chromatography(HPLC).The different cominations were designed with 0,0.5,5,50 mg/L Scutellaria,0,0.13,1.3,13 mg/L Rhubarb,0,0.012,0.12,1.2 mg/L Coptidis,respectively,and then the effect of different drug combinations on PGE_(2) produced by LPS-stimulated HPDLCs was detected by enzyme-linked immunosorbent assay(ELISA).Results The contents of scutellarin,biotin and berberine hydrochloride in the effective parts of Scutellaria,Rhubarb and Coptis were 19.94%,7.50% and 80.87% by HPLC analysis,respectively,and 5 of 32 compatibility groups had significant inhibitory effect on PGE_(2) expression in HPDLCs stimulated by LPS.Five compatibility groups were as following:the compatibility of 5 mg/L ether fraction of Scutellariabaicalensis with 0.12 mg/L Scutellaria water fraction,13 mg/L Rheum ether fraction with 0.012 mg/L Scutellariabaicalensis water fraction,13 mg/L Rheum ether fraction with 0.50 mg/L Scutellariabaicalensis ether fraction,13 mg/L Rheum ether fraction with 5 mg/L Scutellariabaicalensis ether fraction,and 50 mg/L Scutellariabaicalensis ether fraction.Conclusion Five compatibility groups of Scutellaria,Rhubarb and Coptis can inhibit the content of PGE_(2) produced by LPS-stimulated HPDLCs,and provide a basis for the selection of the following experimental dose in vivo.
