摘  要:目的筛选实验性关节炎大鼠滑膜组织中HGF/c-Met信号通路基因表达,探讨实验性关节炎可能的发病机制。方法实验分模型组和正常组,应用Array star大鼠全基因组芯片来检测两组大鼠滑膜组织中mRNAs表达情况,以Fold change值>2.0且P-value<0.05的标准筛选差异表达mRNA,行信号通路分析,筛选与HGF/c-Met信号通路有关的mRNA。结果正常组和模型组共筛选出4547个差异表达mRNA,并获得14条信号通路与c-Met基因表达相关,其中细胞因子-细胞因子受体相互作用信号通路的富集分数最高,共有21个差异基因且均为上调表达,其中c-Met mRNA差异表达倍数为5.19,进而激活下游JAK2/STAT3、PI3K/Akt等信号通路。结论 HGF/c-Met信号通路在实验性关节炎发病过程中发挥重要作用。OBJECTIVE To screen the gene expression of HGF/c-Met signal pathway in synovial tissue of experimental arthritis rats,and explore the possible pathogenesis of experimental arthritis.METHODS The experiment was divided into two groups:Model group and Control group.The expression of mRNAs in the synovial tissue of the two groups was detected with Array star rat whole genome chip.The differential expression mRNA was screened with Fold change value>2.0 and P-value<0.05.The signal pathway analysis was performed to screen the mRNA associated with HGF/c-Met signal pathway.RESULTS 4547 differential expression mRNA were screened out in the control group and the model group,and 14 signal pathways related to c-Met gene expression were obtained.Among them,the cytokine-cytokine receptor interaction signal pathway has the highest enrichment fraction,and 21 differential genes were all up-regulated genes.Among them,the differential expression fold of c-Met mRNA is 5.19,further activate downstream signal pathways,such as JAK2/STAT3,PI3K/Akt and others.CONCLUSION HGF/c-Met signal pathway plays an important role in the pathogenesis of experimental arthritis.
