摘 要:目的测定金线莲不同器官的多糖含量及组织定位。方法微波-超声协同萃取金线莲根、茎、叶中的多糖,并用苯酚-硫酸法测定其含量,用植物组织化学方法定位多糖的贮存部位。结果金线莲茎的多糖含量为4.47%,根为2.12%,叶为0.64%;金线莲中的多糖贮存于根茎的皮层薄壁细胞。结论金线莲多糖贮存于根茎的薄壁细胞,且茎的含量高于根,叶的多糖含量极少;本研究为金线莲高效利用奠定基础。OBJECTIVE To determine the polysaccharide content and locate polysaccharide from different organs of Anoectochilus roxburghii Lind1.METHODS The polysaccharide was extracted from the root,stem and leaf of A.roxburghii by microwave-ultrasonic method and its content was determined by phenol-sulfuric acid method.Phytohistochemistry was used to locate the storage sites of polysaccharides.RESULTS The content of polysaccharide in stem,root,leaf orderly was 4.47%,2.12%,0.64%.The polysaccharide is stored in the cortex parenchyma cells of the rhizome.CONCLUSION The polysaccharide is stored in the parenchyma cells of rhizome,and the polysaccharide content from stem was higher than that from root,while the polysaccharide content from leaf was very little.This study laid a foundation for the efficient utilization of A.roxburghii.
