氨基酸分析仪测定复方甘草酸苷注射液中甘氨酸的含量

摘　　要：目的探究和建立用氨基酸分析仪测定复方甘草酸苷注射液中甘氨酸的含量的方法,完善复方甘草酸苷注射液中甘氨酸含量的监测体系。方法本实验采用氨基酸分析仪法,色谱柱:PEEK材料安玛西亚色谱填料填装色谱柱;检测波长:570 nm;显色剂:茚三酮缓冲液;进样量:10μL;流速:35 mL/h;流动相:缓冲液系统。甘氨酸经离子交换色谱和茚三酮柱后衍生化,根据外标法进行测定。结果甘氨酸在0.3983^1.1949 mg·mL^(-1)浓度范围内线性关系良好(相关R系数大于0.999),精密度RSD为0.31%,平均加样回收率为97.3%,含量测定结果在90.0%^110.0%范围以内。结论该方法的结果准确可行,弥补了原方法的缺陷,并且重现性好、准确度高,注射液中其他成分对测定影响小,因此该方法可用于测定甘氨酸的含量。OBJECTIVE To explore and establish a method to determine the content of glycine in compound glycyrrhizin injection by amino acid analyzer,improve the monitoring system of glycine content.METHODS This expriment used amino acid analyzer,glycine was derived by ion exchange chromatography and ninhydrin column.Chromatographic column:PEEK material amacian chromatographic packing chromatographic column;Detection wavelength:570 nm;Colorant:indenone buffer;Sampling amount:10μL;Flow rate:35 mL/h;Mobile phase:buffer system.According to the external standard method to determine the content of glycine.RESULTS Glycine had good linear correlation between 0.3983 mg·mL-1 and 1.1949 mg·mL-1.correlation coefficient was greater than 0.999 R,the precision RSD was 0.31%,the average recovery rate was 97.3%,and the determination results were within the range of 90.0%-110.0%.CONCLUSION The result of this method is accurate,feasible and simple,it can compensate for the defects of the original method,it has good reproducibility and high precision,the other components in the injection had little effect on the determination.So the method can be used to determine the content of glycine.
