肝郁脾虚方对失眠大鼠血浆和下丘脑胆囊收缩素-8的影响

摘　　要：目的:观察肝郁脾虚方对失眠大鼠血浆和下丘脑胆囊收缩素-8(Cholecystokinin-8,CCK-8)的影响。方法:40只SD雄性大鼠随机取分成5组,正常对照组8只(不参与造模),其余4组均采用睡眠研究领域常用的大鼠慢性夹尾刺激+对氯苯丙氨酸(Para-chlorophenylalanine,PCPA)腹腔注射复合因子+不规则饮食法(逢单日禁食,饮水正常)制造肝郁脾虚失眠模型,造模成功后分为模型组8只、肝郁脾虚方加减颗粒剂高、中、低剂量组各8只,连续灌胃14 d后,使用酶联免疫吸附测定法(ELISA)检测血浆和CCK-8的含量。结果:与正常对照组比较,模型组血浆和下丘脑CCK-8含量均下降,2组比较差异有统计学意义(P<0.05)。与模型组比较,给药中剂量组和低剂量组血浆中CCK-8含量增加,给药高剂量组、中剂量组和低剂量组下丘脑CCK-8含量均增加,比较差异有统计学意义(P<0.05)。结论:肝郁脾虚方具有增加失眠大鼠血浆和下丘脑CCK-8含量的作用。Objective:To observe the effect of Ganyu Pixu formula on plasma and hypothalamus cholecystokinin-8(CCK-8)levels.Methods:Forty healthy male SD rats were randomly divided into 5 groups.The normal group was not modeled.And in the other four groups,chronic tail clamping stimulation,para-chlorophenylalanine(PCPA)intraperitoneal injection and irregular diet(fasting every other day,drinking water normally)were used to create the liver depression,spleen deficiency and insomnia rats.Then the modeled rats were randomly divided into modeled group,high-dose group,medium-dose group,low-dose group and given Ganyu Pixu granules with different doses for 14 days.ELISA method was used to measure the concentration of CCK-8 in plasma and hypothalamus.Results:Compared with the normal control group,CCK-8 in plasma and hypothalamus decreased in modeled group(P<0.05).Compared with modeled group,plasma CCK-8 increased in medium-dose and low-dose groups(P<0.05),hypothalamus CCK-8 increased in high-dose,medium-dose and low-dose groups(P<0.05).Conclusion:The Ganyu Pixu formula could increase the concentration of CCK-8 in insomnic rats plasma and hypothalamus.
