松郁安神方对失眠大鼠基底核多巴胺和腺苷含量的影响

摘　　要：目的:观察松郁安神方对失眠大鼠基底核组织中神经递质多巴胺和腺苷的调节作用。方法:利用失眠大鼠模型,造模成功后分别给予阿普唑仑、松郁安神方(SYF)低剂量和高剂量用药1w。实验过程中观察大鼠的一般情况变化,治疗结束后通过酶联免疫吸附测定(Elisa)方法检测大鼠基底核组织中多巴胺(Dopamine,DA)和腺苷的含量。结果:SYF能够明显改善实验大鼠由失眠引起的症状,作用优于阿普唑仑。同对照组比较,模型组大鼠基底核组织中DA含量显著升高(P<0.01),腺苷含量显著降低(P<0.01)。同模型组比较,阿普唑仑组、SYF高剂量组和SYF低剂量组大鼠基底核组织中DA含量均显著下降(P<0.01),同时腺苷含量显著升高(P<0.01)。结论:SYF能够显著改善实验大鼠由失眠引起的症状,其机制可能与SYF抑制失眠大鼠基底核组织中DA的释放,增加腺苷的释放相关。Objective:To observe the regulating effects of Songyu Anshen prescription(SYF)on the neurotransmitters dopamine(DA)and adenosine in the basal nucleus tissue of insomnia rats.Methods:Alprazolam,low and high dose SYF were given to the insomnia rats for 1 week respectively.The rats general conditions were observed during the experiment.After treatment,the concentrations of DA and adenosine in basal ganglia of the rats were determined by enzyme-linked immunosorbent assay(Elisa).Results:SYF significantly improved the symptoms caused by insomnia in rats,and its effect was superior to that of Alprazolam.Compared with the control group,DA content in the basal ganglia of the model group was significantly increased(P<0.01)and adenosine content was significantly decreased(P<0.01).Compared with the model group,DA content in basal nucleus of rats in alprazolam group,SYF high-dose group and SYF low-dose group decreased significantly(P<0.01),while adenosine content increased significantly(P<0.01).Conclusion:SYF can significantly improve the symptoms caused by insomnia in rats,and its mechanism may be related to the inhibitory effect of SYF on the release of DA and the increase of adenosine in the basal ganglia of insomnia rats.
