摘  要:目的:利用网络药理学方法探讨生脉散治疗快速性心律失常气阴两虚证可能的机制。方法:在TCMSP、Batman-TCM数据库筛选生脉散活性成分,利用Pubchem、Chemspider、 Swiss target prediction数据库预测相关靶点,Genecards数据库预测快速性心律失常靶点,二者取交集得到共同靶点。利用String数据库、Cytoscape软件构建共同靶点PPI网络图并得到Degree值。在DAVID数据库进行GO、KEGG分析,并利用Cytoscape软件构建靶点-通路网络图。结果:本研究收集到生脉散活性成分52个,活性成分相关靶点1672个,快速性心律失常靶点223个,共同靶点46个,以ACE、SRC、NOS3等相互作用最明显,共同靶点以多种方式参与调控蛋白激酶B信号转导、钙离子运输、钾离子转运等91个生物过程,并参与调节钙、cGMP-PKG、VEGF、MAPK等19个信号通路。结论:生脉散可能通过调控钙离子运输、钾离子转运、内皮功能、氧化应激等机制发挥其多活性成分、多靶点、多通路的治疗快速性心律失常气阴两虚证的作用。Objective:To explore the possible mechanisms of Shengmaisan in treating tachyarrhythmia with Qiyin Liangxu through network pharmacology.Methods:The active components of Shengmaisan were screened on the TCMSP and Batman-TCM databases.Its relevant targets were predicted by the Pubchem and Chemspider and Swiss target prediction databases.Genecards database was used to predict the tachyarrhythmia targets.Both targets constructed venn diagrams to obtain common targets.String database and Cytoscape software were used to construct common target PPI network diagram and get the Degree value.The GO and KEGG analysis were performed in the DAVID database,and the target-pathway network map was constructed using Cytoscape software.Results:The study totally collected 52 active components of Shengmaisan,1672 related targets of active components,223 targets of tachyarrhythmia,46 common targets ACE,SRC and NOS3 interacted most obviously.The common targets were involved in regulating 91 biological processes mainly including protein kinase B signaling,calcium ion transport and potassium ion transport in various ways.Its participated in regulating 19 signal pathways mainly including calcium,cGMP-PKG,VEGF and MAPK.Conclusion:Shengmaisan treated tachyarrhythmia with Qiyin Liangxu type by regulating calcium transport,potassium transport,endothelial function,oxidative stress.
