摘 要:目的建立白假丝酵母菌大蜡螟感染模型,研究ngg1对白假丝酵母菌毒力和菌丝形成能力的影响。方法利用白假丝酵母菌野生株SC5314和突变株ngg1-/-分别感染大蜡螟,通过大蜡螟生存曲线比较这2株真菌的毒力,同时采用HE染色、革兰染色和真菌荧光染色的方法观察ngg1基因对白假丝酵母菌生长和菌丝形成能力的影响。结果 SC5314感染后,大蜡螟4 d内全部死亡,而ngg1-/-感染组仍约有80%存活;HE染色和革兰染色提示ngg1-/-突变株在大蜡螟体内几乎被清除掉,真菌荧光染色结果提示SC5314形成大量菌丝,而ngg1-/-突变株无菌丝生成。ngg1-/-突变株的菌丝形成能力明显减弱,并且对大蜡螟的致病性也显著下降。结论 ngg1基因对白假丝酵母菌的毒力和菌丝形成具有重要作用;大蜡螟是可用于研究不同白假丝酵母菌致病性的感染模型。Objective To establish a Candida albicans-Galleria mellonella infection model and reveal the role of ngg1 in the hypha formation and virulence of C. albicans. Methods The G. mellonella were infected with C. albicans SC5314 and ngg1-/-mutants respectively. And each infection group contained 11 randomly chosen G. mellonella larvae. The virulence of these two fungi was examined by survival curve. Besides,HE,Gram-fast stain and fungal fluorescence stain were performed to investigate the influence of ngg1-/-gene on hypha formation of C. albicans. Results The G. mellonella infected with C. albicans SC5314 were all dead while there were still almost 80% of those infected with ngg1-/-mutants survived during four days.The HE and Gram-fast stain results showed that most of ngg1-/-mutants were cleared by the host in the infection model.Besides,there were plenty of hyphae in the G. mellonella infected with C. albicans SC5314 while in those infected with ngg1-/-mutants showed the opposite results by the fungal immunofluorescence stain. The ngg1-/-mutants obviously reduced hypha growth and showed significantly diminished pathogenicity in the G. mellonella infection models. Conclusion This study revealed the important role of ngg1 in hypha formation and virulence of C. albicans. And the G. mellonella infection model is a great choice for the study of pathogenicity of C. albicans.
