摘 要:目的分析ADP-核糖基化样因子(ARL15)基因多态性与延边地区朝鲜族和汉族2型糖尿病(T2DM)及糖尿病肾病(DN)的关联及ARL15与脂联素(PA)及其他临床实验室指标的相关性。方法设计病例对照实验,纳入葡萄糖耐量正常对照组(NGT组)268例,T2DM组393例,DN组90例,选取4个ARL15基因SNP,通过PCR-LDR进行基因分型。使用SHEsis线上软件分析ARL15基因SNP与T2DM及DN的关联。结果在隐性遗传模型中,rs35941位点CC基因型与T2DM患者发生DN的风险相关(P=0.026)。单倍型CCGA和CTAG与高风险DN相关(P<0.05)。在DN患者中,ARL15与PA有相关性(r=0.372,P=0.014)。结论ARL15与PA相关,其SNP位点rs35941基因型CC与高风险DN相关。Objective To analyze the association of ARL15 gene polymorphism with type 2 diabetes mellitus(T2DM)and diabetic nephropathy(DN)of in Yanbian area Korean and Han population,and to explore the correlation between ARL15 and plasma adiponectin(PA)and other clinical laboratory indicators.Methods A case-control experiment was designed,including NGT group(268 cases),T2DM group(393 cases),and DN group(90 cases).Four ARL15 gene SNPs were selected and genotyped by PCR-LDR.SHEsis online software was used to analyze the association of ARL15 gene SNP with T2DM and DN.Results In the recessive genetic model,the CC genotype of rs35941 was associated with an increased risk of DN in T2DM patients(P=0.026).Haplotypes of CCGA and CTAG were associated with increased risk DN(P<0.05).In DN patients,ARL15 was positively correlated with PA(r=0.372,P=0.014).Conclusions ARL15 is related to PA,and its polymorphism SNP locus rs35941 genotype CC is related to DN.
