摘 要:为筛选蒙古黄芪种子萌发前适宜的消毒剂和诱导愈伤组织适宜的培养基,以0.1%HgCl2和2%NaClO处理不同时间,筛选适合蒙古黄芪种子灭菌的最佳方法,并获得无菌苗;分别以无菌苗的下胚轴和叶片为外植体,接种到附加6-BA、2,4-D和NAA不同质量浓度及其组合的培养基中诱导愈伤组织,以筛选适宜培养基,并通过测定其鲜质量,绘制愈伤组织生长曲线。结果显示,蒙古黄芪种子的最佳灭菌方法为0.1%HgCl_(2)灭菌处理10 min;MS+2.0 mg/L 2,4-D+0.5 mg/L 6-BA最适于下胚轴愈伤组织诱导,诱导率达88%,且愈伤组织色绿、致密;MS+2.0 mg/L 6-BA+1.0 mg/L 2,4-D+1.0 mg/L NAA最适于叶片愈伤组织诱导,诱导率达93%,且愈伤组织生长良好,不易褐化。通过绘制愈伤组织生长曲线,结果显示,黄芪下胚轴愈伤组织生长略强于叶片愈伤组织生长,二者均在生长第5天进入对数生长期,第25天进入生长停滞期,说明20~25 d为最适继代周期,可保持其旺盛生长。黄芪种子灭菌处理和愈伤组织诱导培养基的筛选结果可为蒙古黄芪组培快繁提供理论依据。To screen the suitable disinfectant for Astragalus mongholicus seeds before germination and the optimal medium for callus induction,we treated seeds with 0.1%HgCl2 and 2%NaClO for different treatment time to select the best method for the sterilization of Astragalus mongholicus seeds and obtain aseptic seedlings.The hypocotyls and leaves explants of aseptic seedlings were inoculated into MS medium supplemented with 6-BA,2,4-D and NAA at various concentrations and combinations for callus induction to choose the best medium.The callus growth curve was drawn by measuring the fresh weight of the callus.The results showed that the best method to sterilize Astragalus mongholicus seeds was 0.1%HgCl_(2)for 10 min.The optimal medium for callus induction of hypocotyls explants was MS+2.0 mg/L 2,4-D+0.5 mg/L 6-BA and the induction rates were 88%,and the callus was green and dense.The optimal medium for callus induction of leaves explants was MS+2.0 mg/L 6-BA+1.0 mg/L 2,4-D+1.0 mg/L NAA and the induction rates reached 93%,and the callus grown well and was not easy to brown.The callus growth curve showed that the callus growth entered the log phase on the 5th day and entered the stationary phase on the 25th day.The optimum subculture period was within 20-25 d,which could maintain its vigorous growth.In addition,the callus growth of hypocotyl explants was slightly heavier than that of leaf explants.The sterilization of seeds and the selection results of callus induction medium will provide a theoretical basis for tissue culture and rapid propagation of Astragalus mongholicus.
