摘 要:利用小麦-中间偃麦草部分双二倍体TAI7045与普通小麦晋太170进行亚远缘杂交选育的小偃麦新种质CH366高抗小麦条锈菌条中32、条中33和条中34号生理小种,为了追踪其抗条锈病基因来源,对CH366及其亲本接种条锈菌生理小种,并对CH366进行了荧光原位杂交(FISH)鉴定。结果表明,CH366和TAI7045对条锈病表现为近免疫和免疫,其小麦亲本晋太170则表现为高感,证明CH366的条锈病抗性来源于TAI7045;FISH鉴定表明,CH366中含有44条染色体,其中,42条来源于晋太170,另2条来源于TAI7045的中间偃麦草染色体。CH366是一个小麦-中间偃麦草二体异附加系,可作为小麦抗条锈病遗传改良的中间亲本。Thinopyrum intermedium,one of the important gene sources for wheat genetic improvement,can improve significantly the disease resistance of cultivated wheat by introducing its resistance gene into wheat backgroud.CH366,a novel germplasm with resistance to stripe rust of Pst CYR32,CYR33 and CYR34,derived from sub-wide cross of wheat-Thinopyrum intermedium partial diploid TAI7045 and common wheat Jintai 170.To trace the origin of stripe rust resistance genes,the resistance of CH366 and its parents were identified by artificial inoculation with stripe rust physiological races,and CH366 was identified by fluorescence in situ hybridization(FISH).The results showed that CH366 and TAI7045 showed near immunity and immunity while Jintai 170 showed high sensitivity to stripe rust.It suggested that the resistance of CH366 might come from TAI7045.FISH results showed that CH366 contained 44 chromosomes,of which 42 were derived from Jintai 170 and the other 2 from Thinopyrum intermedium chromosomes of TAI7045.As a result,CH366 is a wheat-Thinopyrum intermedius disomic alien additional line,which can be used as an intermediate parent for genetic improvement of wheat resistance to stripe rust.
