摘  要:建立了UFLC-MS/MS法测定大鼠左旋多巴的血浆浓度。色谱柱选用Phenomenex Gemini C_(18)柱(2.0 mm×100 mm,5μm),流动相A为0.2%甲酸溶液,B为乙腈,流速为0.3 ml/min,进样量为10μl;柱温为40℃。结果显示,在100~6000 ng/ml内,左旋多巴的校准曲线呈线性,定量下限为100 ng/ml,含左旋多巴的血浆平均回收率为98.87%~101.34%。方法专属性好,精密度高,为左旋多巴血浆样品的定量分析提供参考。An UFLC-MS/MS method was established to determine the plasma concentration of levodopa in rats.An Phenomenex Gemini C_(18)column(2.0 mm×100 mm,5μm)was used,with the mobile phase A of 0.2%formic acid solution,and the mobile phase B of acetonitrile.The flow rate was 0.3 ml/min,the injection volume was 10 ml,and the column temperature was 40℃.The results showed that it was linear for levodopa calibration curve in the range of 100-6000 ng/ml.The lower limit of quantification was 100 ng/ml and the average recovery rate of plasma containing levodopa was 98.87%-101.34%.The method is specific and precise,which provides a reference for the quantitative analysis of levodopa plasma samples.
