摘  要:目的分析用基质辅助激光解吸附电离飞行时间质谱(MALDI-TOF MS)与16S r RNA鉴定副溶血弧菌(VP)的准确性、快速性以及经济性。方法用Autof ms1000质谱仪鉴定37株从沈阳市疾病预防控制中心获得的弧菌,获得鉴定结果和弧菌图谱,同时做聚类分析与16S rRNA测序。结果37株弧菌质谱鉴定后得出编号1~9、11~37为副溶血弧菌,10为霍乱弧菌,鉴定结果种属水平可信。16S rRNA鉴定出1~9、10、11~37均为弧菌属,第一匹配和第二匹配相似率差异<0.8%。结论Autof ms1000在鉴定菌株方面经济快速,鉴定37株弧菌准确性在属水平上达到100%,种水平上达到97.3%,在属水平上与16S rRNA测序高度一致。Objective To analyze the accuracy,rapidness and economy of identification of Vibrio parahaemolyticus(VP)by matrixassisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS)and 16 S rRNA.Methods Autof ms1000 spectrometer was used to identify 37 strains of Vibrio obtained from Shenyang Center for Disease Control and Prevention,to identification results and Vibrio atlas.And the cluster analysis and 16 S r RNA sequencing were performed.Results In 37 strains of Vibrio,it was identified No.1-9 and 11-37 as Vibrio parahaemolyticus and No.10 as Vibrio cholerae by mass spectrometry.The identification results are reliable in species level.No.1-9,10 and 11-37 were identified as Vibrio by 16 S r RNA.Difference in similarity rate between first match and second match was<0.8%.Conclusion Autof ms 1000 is economical and rapid in identifying strains.The accuracy of identifying 37 strains of Vibrio reaches 100%at the genus level and 97.3%at the species level,which is highly consistent with 16 S rRNA sequencing at the genus level.
