摘 要:目的考察氧化三甲胺(TMAO)对血管内皮细胞炎症以及肠上皮细胞炎症的影响。方法采用不同浓度的TMAO处理HUVECs细胞,考察不同时间点的炎症因子(ICAM-1、VCAM-1)表达情况;采用不同浓度的TMAO处理HT29细胞,考察不同时间点的炎症因子(IL-1β、IL-8和IL-23)表达情况;考察TMAO对紧密连接蛋白(ZO-1和Occludin-1)表达的影响;小鼠体内实验分为对照组(n=3)和TMAO组(n=3),观察TMAO在各组织中的分布。结果TMAO增强TNF-α诱导的人脐静脉内皮细胞黏附因子表达;TMAO可以促进HT29细胞IL-8、IL-1β炎症因子的表达;TMAO在处理细胞6 h可促进紧密连接蛋白的表达;TMAO在小鼠各组织中的分布以肺脏最多。结论TMAO增强血管内皮细胞炎症因子表达,促进肠上皮细胞炎症和紧密连接蛋白表达。Objective To explore the effects of trimethylamine N-oxide(TMAO)on vascular endothelial cell and intestinal epithelial cell inflammation.Methods Human umbilical vein cells(HUVECs)were treated with different concentrations of TMAO to observe the expression of inflammatory cytokines(ICAM-1,VCAM-1)at different time points.HT29 cells were treated with different concentrations of TMAO and the expression levels of inflammatory cytokines(IL-1β,IL-8,and IL-23)at different time points were observed.The effects of TMAO on the expression of tight junction proteins(ZO-1 and Occludin-1)were examined.We also examined the distribution of TMAO in tissue in vivo using subdermal injection of TMAO.Results TMAO synergistically enhanced TNF-α-induced inflammatory cytokine expression in HUVECs.TMAO promoted the expression of IL-8 and IL-1βinflammatory cytokines in HT29 cells,and promoted the expression of tight junction proteins in epithelial cells.Tissue distribution study revealed that exogenous TMAO was more likely targeted to the lung.Conclusion TMAO enhances the expression of inflammatory cytokines in vascular endothelial cells,and promotes intestinal epithelial cell inflammation and expression of tight junction proteins.
