摘 要:试验旨在研究由金银花、熟地等组成的中草药复方制剂对感染IBDV青脚麻鸡脾脏显微结构、抗氧化功能、细胞因子表达、淋巴细胞凋亡及TLR3/NFκB信号通路的影响。随机将500羽健康28日龄未接种法氏囊疫苗的青脚麻鸡分为5组,对照组、攻毒组和试验Ⅰ—Ⅲ组,3个试验组日粮中分别对应给予0.5%、1%和1.5%的中草药复方制剂。除对照组外的其余组均在30日龄通过点眼人工感染IBDV,试验期7 d。IBDV攻毒3 d后,比较对照组,攻毒组青脚麻鸡脾指数,脾脏MDA含量,NFκB和TLR3基因表达量(mRNA),Caspase-3基因和蛋白表达量及其阳性细胞数均呈现显著增加(P<0.05),而T-SOD与T-AOC活性和脾脏中IFN-γ与IL-2含量呈现显著减少(P<0.05)。比较攻毒组,试验Ⅱ组青脚麻鸡脾脏MDA含量、Caspase-3基因和蛋白表达量均呈现明显降低(P<0.05),而脾脏中IL-2和IFN-γ含量及T-SOD和T-AOC活性则明显提升(P<0.05)。IBDV攻毒7 d后,比较对照组,攻毒组青脚麻鸡脾脏的器官指数、Caspase-3 mRNA和蛋白表达量及其单位面积阳性细胞数同样呈现明显增加(P<0.05),同时NFκB和TLR3基因表达量也出现明显增加(P<0.05),而脾脏中细胞因子IL-2及GSH-Px和T-SOD的含量或活性增明显下降(P<0.05)。比较攻毒组,试验Ⅱ组青脚麻鸡脾指数、MDA含量、Caspase-3和NFκB基因表达量均呈现明显降低(P<0.05),而T-AOC、T-SOD活性及上述细胞因子含量则明显升高(P<0.05)。感染3 d和7 d后,显微观察可见试验Ⅱ组青脚麻鸡的脾小结和动脉周围淋巴鞘数量增加,脾脏结构更清晰,与对照组相比基本相同,与攻毒组相比,脾小结和动脉周围淋巴鞘体积明显降低。日粮中添加1%的中草药复方制剂对感染IBDV青脚麻鸡脾脏显微结构、免疫抗氧化功能和TLR3/NFκB信号通路具有明显促进作用,在一定程度上抑制了脾脏淋巴细胞凋亡。To study the effects of Chinese herbal medicine composed of honeysuckle and cooked ground antioxidant function, expression of cytokines, apoptosis of lymphocytes and TLR3/NFκB signal pathway of green feet chicken infected with IBDV. 500 healthy 28-day-old broilers without bursal vaccine were divided into five groups: control group, attack group and test Ⅰ-Ⅲ group. The test group Ⅰ, Ⅱ and Ⅲ were supplemented with 0.5%, 1.0% and 1.5%compound Chinese herbal medicine respectively. Except the control group, all the other groups were infected with IBDV artificially by eye spot at the age of 30 days, the test lasted 7 days. After 3 days of IBDV infection, compared with the control group, the spleen index, MDA content, NFκB and TLR3 gene expression(mRNA), Caspase-3 gene and protein expression and the number of positive cells were significantly increased(P<0.05), while the activity of T-SOD and T-AOC and the content of IFN-γ and IL-2 in spleen were significantly decreased(P<0.05). Compared with the attack group, the MDA content, Caspase-3 gene and protein expression in spleen of test group Ⅱ decreased significantly(P<0.05), while the IL-2 and IFN-γ content, T-SOD and T-AOC activity in spleen increased significantly(P<0.05). After 7 days of IBDV infection, the organ index, Caspase-3 mRNA and protein expression and the number of positive cells per unit area in spleen of the control group were also significantly increased(P<0.05), while the expression of NFκB and TLR3 genes were also significantly in-creased(P<0.05), but the content or activity of IL-2, GSH-PX and T-SOD in spleen were significantly de-creased(P<0.05). Compared with the attack test group, the spleen index, MDA content, Caspase-3 and NFκB gene expression in group Ⅱ decreased significantly(P<0.05), while T-AOC, T-SOD activity and the above cytokines content increased significant(P<0.05). Three days and seven days after IBDV infection, the number of spleen nodule and periarterial lymphosheath increased, the structure of spleen was more clear
