摘 要:目的观察miR-137在急性心肌梗死(AMI)患者和人脐静脉内皮细胞(HUVECs)中的作用,探索AMI诊断和治疗的新方法。方法利用生物信息学工具TargetScan预测microRNA-137(miR-137)与Kruppel样因子2(KLF2)的相互作用,并通过荧光素酶报告实验进行验证。用CCK-8法检测miR-137对HUVECs活力的影响。采用实时荧光定量PCR检测血清miR-137的表达,并分析其与心肌损伤、内皮损伤标志物和促炎细胞因子的相关性。建立受试者工作特征曲线(ROC),评价miR-137在AMI患者中的诊断价值。结果 miR-137作为KLF2的直接调节因子,可抑制HUVECs的增殖能力。血清miR-137在AMI患者中表达水平升高,与患者心肌损伤标志物、内皮损伤标志物以及前炎症因子水平呈正相关。根据miR-137水平建立的ROC曲线下面积为0.949,提示miR-137对AMI有较高的诊断准确性。结论 miR-137可能通过靶向KLF2参与AMI的心肌损伤、内皮损伤和炎症反应,miR-137表达水平的调控将为探究AMI治疗新方法提供思路。Objective The purpose of this study was to analyze the function of miR-137 in acute myocardial infarction(AMI)patients and human umbilical vein endothelial cells(HUVECs),and explore novel diagnostic and therapeutic methods for AMI.Methods The interaction between miR-137 and KLF2 was evaluated using bioinformatics analysis with TargetScan and a luciferase reporter assay.The effect of miR-137 on HUVEC viability was assessed using CCK-8 assay.The serum expression of miR-137 was examined using qRT-PCR,and its correlation with markers of myocardial and endothelial injury and pro-inflammatory cytokines was analyzed.The receiver operating characteristics(ROC)curve as constructed to evaluate the diagnostic value of miR-137 in AMI patients.Results miR-137,as a direct regulator of KLF2,could inhibit HUVEC proliferation.Serum miR-137 expression was elevated in AMI patients and positively correlated with markers of myocardial and endothelial injury and pro-inflammatory cytokines.The area under the ROC curve based on serum miR-137 levels was 0.949,indicating that miR-137 had relatively high diagnostic accuracy in AMI.Conclusion The findings of this study revealed that miR-137 is involved in the myocardial and endothelial injury and inflammation by targeting KLF2.In addition,the overexpression of miR-137 in AMI patients has certain diagnostic accuracy,which may be a novel candidate diagnostic biomarker for AMI.
