摘  要:目的探讨Wnt5a在家兔背根神经节损伤模型中的表达变化及神经根病中Wnt5a与TNF-α的关系。方法选取家兔96只,随机分为4组:空白对照组(A组)、穿刺组(B组)、穿刺+生理盐水组(C组)、穿刺+依那西普组(D组)。各组术后1d,3d,5d,7d分别行RT-PCR和免疫印迹法检测。结果术后1d,3d,5d,7d,B、C组Wnt5a mRNA相对表达量和蛋白表达显著升高,且高于A、D组(P<0.05),组间比较差异无统计学意义(P>0.05);D组术后1d、3d时Wnt5a mRNA相对表达量及蛋白表达升高,而且高于A组,低于B和C组(P<0.05),术后5d、7d时Wnt5a mRNA相对表达量及蛋白表达量降低,仍高于A组(P<0.05)。结论有椎间盘退变突出等压迫因素压迫背根神经节,致背根神经节受损时,Wnt5a表达量增多,抑制TNF-α后表达量减少,可能激活Wnt5a介导的负反馈机制,使得表达量进一步减少。Objective The expression of Wnt5 a in dorsal root ganglion injury model of rabbits and the relationship between Wnt5 a and TNF-αin radiculopathy.Methods Ninty-six rabbits were randomly divided into 4 groups:blank control group(group A),puncture group(group B),puncture+normal saline group(Group C),puncture+etanercept group(Group D).RT-PCR and Western blot were performed on the 1 st,3 rd,5 th and 7 th day after operation.Results The relative expression of Wnt5 a mRNA and protein in group B and C were significantly higher than those in group A and group D(P<0.05).On the 1 st,3 rd,5 th,7 th day after operation,but there was no significant difference between groups(P>0.05);the relative expression of Wnt5 a mRNA and protein in group D was higher than that in group A,lower than that in group B and C(P<0.05).On the 1 st,3 rd day after operation.The relative expression of mRNA and protein in group A was lower than that in group A(P<0.05).On the 5 th,7 th day after operation.Conclusion When the dorsal root ganglion is compressed by compression factors such as intervertebral disc degeneration and protrusion,the dorsal root ganglion is damaged,the expression of Wnt5 a increased and decreased after inhibition of TNF-α,which may activate the negative feedback mechanism mediated by Wnt5 a.
