摘 要:目的建立一测多评法同时测定山苦荬Ixeridium chinense(Thunb.)Tzvel.中绿原酸、咖啡酸、木犀草苷的含量。方法山苦荬70%甲醇提取物的分析采用CAPCELLPAK TYPEⅡSIZE C_(18)柱(4.6 mm×250 mm,5μm);流动相甲醇⁃0.2%磷酸溶液,梯度洗脱;体积流量1.0 mL/min;柱温25℃;测定波长330 nm,以绿原酸为内标,计算其他2种成分的相对校正因子,再测定其含量。结果绿原酸、咖啡酸、木犀草苷分别在13.389~53.556μg/mL(r=0.9960)、8.070~32.280μg/mL(r=0.9971)、61.356~254.424μg/mL(r=0.9940)范围内线性关系良好,平均回收率分别为99.15%、99.01%、101.48%,RSD分别为2.3%、2.4%、2.6%。一测多评法所得结果接近于外标法。结论该方法准确稳定,重复性好,可用于山苦荬的质量控制。AIM To establish a quantitative analysis of multi⁃components by single mark(QAMS)method for the simultaneous content determination of chlorogenic acid,caffeic acid and luteolin in Ixeridium chinense(Thunb.)Tzvel..METHODS The analysis of 70%methanol extract of I.chinense was performed on a 25℃thermostatic CAPCELLPAK TYPEⅡSIZE C_(18) column(4.6 mm×250 mm,5μm),with the mobile phase comprising of methanol-0.2%phosphoric acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 330 nm.With chlorogenic acid as an internal standard,the relative correction factors of the other two constituents were calculated,followed by the determination of their contents.RESULTS Chlorogenic acid,caffeic acid and luteolin showed good linear relationships within the ranges of 13.389-53.556μg/mL(r=0.9960),8.070-32.280μg/mL(r=0.9971),61.356-254.424μg/mL(r=0.9940),whose average recoveries were 99.15%,99.01%,101.48%with the RSDs of 2.3%,2.4%,2.6%,respectively.The results obtained by QAMS approximated those obtained by external standard method.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of I.chinense.
