摘 要:目的:探究分心木提取物预处理对大鼠肝缺血-再灌注损伤(HIRI)的保护作用。方法:将30只SD雄性大鼠随机分为空白组(S组)、缺血-再灌注组(I/R组)和给药组(DJ组),每组10只。DJ组进行分心木提取物给药干预,S组、I/R组给予等量0.9%氯化钠溶液。14d后,建立大鼠HIRI模型,S组除外。24 h后取材检测缺血组织肝功能及生化指标,并通过苏木精-伊红染色观察肝组织病理变化。结果:与S组比较,I/R组肝组织中AST、ALT和XOD活力升高(P<0.01),MDA、Hcy和NO的含量均升高(P<0.05或P<0.01),SOD和GSH-Px水平降低(P<0.01),荧光显微镜下观察I/R组大鼠肝细胞明显变性、坏死,而经给药预处理的DJ组上述情况均有不同程度改善。结论:分心木乙酸乙酯提取物预处理对大鼠HIRI具有一定的抗氧化保护作用。Objective:To explore the protective effects of Diaphragma juglandis Fructus extract pretreatment on rat hepatic ischemiareperfusion injury(HIRI).Methods:30 SD male rats were randomly divided into the sham operation group(S group),the ischemiareperfusion group(I/R group)and the drug treatment group(DJ group),with 10 rats in each group.The DJ group was given the administration intervention of the extracts of Diaphragma juglandis Fructus,and the S,I/R groups were given the same volume of 0.9%sodium chloride solution.After 14 days,a rat HIRI model was established,except for the S group.After 24 hours,samples were taken to detect the liver function and biochemical indexes of ischemic tissue,and the pathological changes of liver tissue were observed by hematoxylin-eosin staining.Results:Compared with S group,the activities of AST,ALT and XOD in liver tissues of I/R group increased(P<0.01),the contents of MDA,Hey and NO increased(P<0.05 or P<0.01),while SOD and GSH-Px levels decreased(P<0.01).Observed under a fluorescence microscope,the rat liver cells in the I/R group were obviously degenerated and necrotic,while the abovementioned conditions in the DJ group pretreated by the administration were improved to varying degrees.Conclusion:The pretreatment of the ethyl acetate extract of Diaphragma juglandis Fructus has a certain antioxidant effects on HIRI in rats.
