甲基红褪色光度法测定司帕沙星含量的研究

摘  要:建立了褪色光度法测定司帕沙星含量的方法。室温下,在pH=3.5的HAc-NaAc缓冲体系中,司帕沙星与甲基红溶液反应10 min形成配位比1∶1的荷移络合物,在波长553 nm处有最大的褪色吸收峰。在此波长下,司帕沙星的浓度c在1.00×10-6~3.00×10-6 mol/L遵守朗伯比尔定律,线性方程为A=0.14936 c+0.09909,相关系数r=0.997,检出限D=6.00×10-7 mol/L。在该浓度范围内,表观摩尔吸光系数K=2.10×105 L/(mol·cm);样品平均回收率为101.5%,RSD为1.52%,可用于准确测定药剂中司帕沙星的含量。A new method for the determination of Sparfloxacin content by fading photometry was developed.In the Sodium Acetate buffer system of pH 3.5,at room temperature,Sparfloxacin reacts shows with Methyl Red solution to form an charge transfer complex with a coordination ratio of 1:1 at 10 min,and has the largest fading absorption peak at 553 nm.At this wavelength,Sparfloxacin had a good internal relationship in the range of 1.00×10-6~3.00×10-6mol/L concentration,liner equation for A=0.14936 c+0.09909,r=0.997,the detection limit is 6.00×10-7 mol/L.In this range,the apparent molar absorption coefficient is 2.10×105 L/(mol·cm);the average recovery rate is 101.5%,RSD%1.52.The method can be used for the determination of Sparfloxacin in medicine.
