摘  要:灌胃大黄建立小鼠腹泻模型,通过测定小鼠血清中和结肠组织中水通道蛋白3(aquaporin 3,AQP3)、水通道蛋白4(aquaporin 4,AQP4)及血管活性肠肽(vasoactive intestine polypeptide, VIP)的含量变化,探究大黄致泻下的作用机制。选取40只健康昆明种小鼠,随机分为4组,每组10只,B、C、D组灌胃不同浓度的大黄混悬液(B组1 g/kg、C组2 g/kg、D组4 g/kg),A组给予等体积的生理盐水,连续灌胃5 d。采用ELISA、免疫组织化学法和荧光定量PCR检测血清及结肠组织中AQP3、AQP4、VIP的含量及表达情况。结果显示,大黄组小鼠体重显著减轻,采食量减少,随给药剂量的增加,腹泻逐渐加剧,C组、D组小鼠血清中AQP3、AQP4含量较A组显著降低,结肠组织中AQP3、AQP4与VIP表达量较A组显著降低(P<0.05),与B组无明显差异。结果表明,大黄可通过降低AQP3、AQP4表达量来影响结肠对水的吸收,增加肠道含水量,通过降低VIP表达量使肠蠕动增加,从而使小鼠排便增加,引起腹泻。In order to investigate the mechanism of Rheum palmatum L.induced diarrhea,a model of diarrhea in mice was established by gavage of Rheum palmatum L.,and the contents of aquaporin 3(AQP3),aquaporin 4(AQP4)and vasoactive intestine polypeptide(VIP)in the serum and colon tissue of mice were determined.Forty healthy Kunming mice were selected and randomly divided into 4 groups of 10 mice each.Groups B,C,and D were administered with Rheum palmatum L.suspensions of different concentrations(group 1 g/kg,group C 2 g/kg,Group D(4 g/kg))intragastrically for 5 days,group A was given an equal volume of normal saline.The contents and expressions of AQP3,AQP4,VIP in serum and colon tissue were detected by ELISA,immunohistochemistry and fluorescent quantitative PCR.The results showed that the mice in the Rheum palmatum L.groups significantly reduced body weight and feed intake.With the increase of the dose,diarrhea gradually intensified.The levels of AQP3 and AQP4 in the serum of the mice in the C and D groups were significantly lower than those in the A group,and the expression levels of AQP3,AQP4 and VIP in the colon tissue were significantly lower than those in group A(P<0.05),and there was no significant difference from group B.The results showed that Rheum palmatum L.can affect the colon's absorption of water by reducing the expressions of AQP3 and AQP4,increasing the intestinal water content and the bowel motility by reducing the expression of VIP,thereby increasing bowel movements in mice and causing diarrhea.
