摘 要:为了解近年鹅星状病毒的流行情况,通过鹅胚接种、鹅胚半数致死量测定(ELD_(50))、RT-PCR检测和基因组序列测定等方法,从2019年山东某养鹅场雏鹅疑似星状病毒引起鹅内脏痛风病的病料中分离出1株新型鹅星状病毒,命名为AstV/Goose/SD776/2019。该分离株能引起鹅胚典型的临床症状,病毒无血凝活性,ELD_(50)为10^(-4.0)/0.2 mL,动物回归试验中死亡的雏鹅可以复制出与临床自然感染相似的病变。用RT-PCR对分离株全基因组序列分析结果显示,分离株的3个ORF和与新型鹅星状病毒毒株(JSHA、SD01、SDPY、GD)的核苷酸和氨基酸同源性较高(99.2%以上);与能导致雏鹅肠炎的鹅星状病毒(FLX)及其他种属禽源星状病毒同源性相对较低,遗传进化分析结果显示,分离株在进化树上与4株新型鹅星状病毒的亲缘关系接近,表明分离毒株为新型鹅星状病毒。To better understand the prevalence of goose astrovirus in recent years,the methods of goose embryo inoculation,ELD50,RT-PCR detection and genome sequencing were adopted in this experiment.A strain of goose astrovirus was isolated from the geese of a goose farm in Shandong province in 2019 that were clinically suspected to be gout caused by astrovirus,named AstV/Goose/SD776/2019.The results showed that this strain could cause typical clinical symptoms of goose embryo.The virus had no hemagglutination activity,and ELD 50 was 10^(-4.0)/0.2 mL.Goslings that died in animal regression tests could replicate lesions similar to clinical natural infections.RT-PCR analysis of the genome sequence of the isolate showed that the nucleotide and amino acid homologies of three ORF of the isolate with the new goose astrovirus strains(JSHA,SD01,SDPY,GD)were higher than 99.2%.It had relatively low homology with goose astrovirus(FLX)and other species of avian astrovirus that can cause gosling enteritis.The results of genetic evolution analysis showed that the isolate is most closely related to the four new goose astrovirus strains in the evolutionary tree and belongs to the same branch,indicating that the isolated strain belongs to the new goose astrovirus.
