摘 要:目的:探讨加味当归贝母苦参丸对慢性非细菌性前列腺炎(CNP)大鼠前列腺组织炎症及脊髓氧化应激的影响。方法:取2月龄雄性SD大鼠60只,随机选取10只作为正常对照组,另50只采用前列腺腹叶注射完全弗氏佐剂法制作CNP模。将成模大鼠随机分为加味当归贝母苦参丸低(3 g/kg)、中(6 g/kg)、高剂量组(12 g/kg),前列倍喜组(1.6 g/kg),模型组,各组均灌胃给药,1次/天,连续药物干预4周。取各组大鼠前列腺组织,光镜下观察各组大鼠前列腺组织形态学变化,酶联免疫吸附法检测脊髓组织及神经节8-epi PGF2a(8-异前列腺素)物质水平,采用黄嘌呤氧化酶法测定T-SOD(总超氧化物歧化酶)活力。结果:①前列腺病理学观察:模型组前列腺组织炎症明显,前列腺上皮细胞细胞核明显增大,形状不规则,排列紊乱,间质炎细胞浸润明显,水肿;加味当归贝母苦参丸及前列贝喜胶囊组的病理损伤均有不同程度的减轻。②脊髓氧化应激:与正常对照组相比,模型组及低剂量组8-epi PGF2a水平明显增高,T-SOD活力降低;差异有统计学意义(P<0.01);加味当归贝母苦参丸中、高剂量及前列腺倍喜胶囊组8-epi PGF2a水平明显低于模型组,T-SOD活力高于模型组,差异有统计学意义(P<0.01,P<0.05)。结论:加味当归贝母苦参丸能减轻前列腺组织的病理损伤,改善模型大鼠脊髓氧化应激,这可能是加味当归贝母苦参丸改善前列腺炎患者症状的分子机制之一。Objective:To investigate the effects of Jiawei Danggui Beimu kushen Pill on inflammatory of prostate tissue and oxidative stress of spinal cord in rats with chronic nonbacterial prostatitis(CNP).Methods:602-month-old male SD rats were randomly selected as control group and 50 were molded with complete freund’s adjuvant injected in the ventral prostate lobe,after the models were successfully established,the molding rats were randomly divided into Jiawei Danggui Beimu Kushen Pill low(3 g/kg),medium(6 g/kg)and high dose(12 g/kg)groups,Qianlie Beixi Capsule group(1.6 g/kg)and model group,each group was given corresponding medicine or saline by gavage once a day for 4 weeks.The prostate tissues of rats in each group were taken and the changes of prostate histomorphology of rats in each group were observed under light microscope.The level of substance 8-epi PGF2a in spinal cord tissues and dorsal root ganglion(DRG)was detected by enzyme-linked immunoadsorption method,and the activity of T-SOD was determined by xanthine oxidase method.Results:①Prostate histomorphology:in the model group,the prostate tissue was obviously inflamed,the prostatic epithelial cell nucleus was obviously enlarged,the shape was irregular,the arrangement was disordered,the interstitial inflammatory cell infiltration was obvious,edema;The pathological damage of Jiawei Danggui Beimu Kushen Pill and Qianlie Beixi Capsule were improved to different degrees.②Spinal cord oxidative stress:compared with the normal control group,the level of 8-epi PGF2a in the prostatic tissue of the model group and the low-dose group were increased significantly,and the activity of T-SOD was decreased,the difference was statistically significant(P<0.01);The level of 8-epi PGF2a in the medium and high dose Jiawei Danggui Beimu Kushen Pill and Qianlie Beixi Capsule groups was significantly lower than that in the model group,and the activity of T-SOD was higher than that in the model group,the differences were statistically significant(P<0.01,P<0.05).Conclusion:Jiawei
