摘  要:目的:应用网络药理学方法探究那如三味丸治疗类风湿关节炎(Rheumatoid arthritis,RA)的作用机制。方法:通过TCMSP数据库和SwissTarget Prediction数据库获得那如三味丸单味药的所有成分及靶点信息,通过DisGeNET数据库获取RA靶点,筛选药物及疾病靶点交集,获得那如三味丸治疗RA的潜在靶点。对靶点进行蛋白-蛋白相互作用分析、网络分析及Hub节点选择,明确靶点在治疗RA中的意义。结果:得到那如三味丸治疗RA靶点106个,那如三味丸主要参与34个生物功能,31个生物过程,6个免疫过程,调节磷脂酰肌醇3激酶-蛋白激酶B(Phosphatidylinositol 3 kinase-protein kinase B,PI3K-Akt)、趋化因子、低氧诱导因子-1(Hypoxia inducible factor-1,HIF-1)等与RA密切相关的信号通路22条,确定那如三味丸治疗RA的关键靶点为丝裂原活化蛋白激酶1(Mitogen-activated protein kinase1,MAPK1),丝裂原活化蛋白激酶3(Mitogen-activated protein kinase3,MAPK3)。结论:通过网络药理分析预测到那如三味丸治疗RA的靶点及其参与的生物学过程,反映出蒙药多组分、多靶点、多途径的特点,为后续研究提供了理论基础。Objective:To explore the mechanism of Naru Sanwei Pills in the treatment of rheumatoid arthritis(RA)by using network pharmacology methods.Methods:All the chemicals and targets information of Naru Sanwei Pill ingredients were obtained through TCMSP database and SwissTarget Prediction database,RA targets were obtained through DisGeNET database,the intersection of drugs and disease targets were screened,and the potential target of Naru Sanwei Pill in treating RA were obtained.Protein-protein interaction analysis,network analysis and Hub node selection were performed on the target to clarify the significance of the target in the treatment of RA.Results:106 targets of Naru Sanwei Pills for the treatment of RA were obtained.Naru Sanwei Pills were mainly involved with 34 biological functions,31 biological processes and 6 immune processes.22 signal pathways closely related to RA,such as phosphatidylinositol 3-kinase-protein kinase B(PI3K-Akt),chemokines,and hypoxia-inducible factor-1(HIF-1)were regulated to determine the key targets of Naru Sanwei pills in the treatment of RA.The points were mitogen-activated protein kinase 1(MAPK1)and mitogen-activated protein kinase 3(MAPK3).Conclusion:Through network pharmacological analysis,the targets of Naru Sanwei Pill in treating RA and the biological process involved in it were predicted,reflecting the characteristics of multi-component,multi-target,and multi-pathway of Mongolian medicine,which provides a theoretical basis for follow-up research.
