摘 要:目的探讨两固汤加减对促排卵模型小鼠种植窗期子宫内膜容受性的影响及可能作用机制。方法144只雌性ICR小鼠随机分为自然周期组、促排卵组、黄体酮组、两固汤加减组,每组36只。促排卵组依据体重给予生理盐水0.2ml/10g灌胃,黄体酮组给予黄体酮0.4mg/10g灌胃,两固汤加减组给予两固汤加减0.3mg/10g灌胃,每日1次,连续15天。连续灌胃的后9天,促排卵组、黄体酮组、两固汤加减组雌鼠采用孕马血清+醋酸曲普瑞林注射液法建立促排卵小鼠模型,自然周期组雌鼠腹腔注射等体积生理盐水。最后一次注射后各组小鼠雌雄2∶1合笼。合笼第2天早上观察雌鼠阴栓情况、胚胎着床数,观察种植窗期孕鼠子宫内膜组织学变化特点,测量子宫内膜螺旋动脉数量。分别于交配日后第2、3、4、5、6、7日处死小鼠各6只,检测小鼠子宫内膜巨噬细胞炎症蛋白1β(MIP1β)、CD244、子宫自然杀伤细胞(uNK)蛋白表达。结果与自然周期组比较,促排卵组交配日后第2天胚胎着床数显著减少,交配日后第4天子宫内膜螺旋动脉数量减少(P<0.05)。与促排卵组比较,黄体酮组及两固汤加减组小鼠胚胎着床数、子宫内膜螺旋动脉数数量增加(P<0.05)。第4、5天,促排卵组CD244、uNK蛋白表达较自然周期组降低,而黄体酮组和两固汤加减组较促排卵组升高,且两固汤加减组在第4天高于黄体酮组和自然周期组(P<0.05)。结论两固汤加减可改善促排卵模型小鼠种植窗期子宫内膜容受性,促进胚胎着床,其机制可能通过纠正子宫内膜uNK、CD244蛋白表达时序紊乱有关。Objective To explore the effect of Modified Lianggu Decoction(两固汤加减)on endometrial receptivity during the implantation window of ovulation induction model mice,and the underlying mechanism.Methods Totally 144 female ICR mice were randomly divided into natural cycle group,ovulation induction group,progesterone group,and Modified Lianggu Decoction group,with 36 mice in each group.Normal saline 0.2 ml/10 g(body weight),progesterone 0.4 mg/10 g,and Modified Lianggu Decoction 0.3 mg/10 g were administered by gavage,once daily,for 15 days in ovulation induction group,progesterone group and Modified Lianggu Decoction group,respectively.After nine days of gavage,pregnant mare serum and triptorelin acetate injections were used to establish a model of ovulation induction in female mice of ovulation induction group,progesterone group and Modified Lianggu Decoction group,while equal volume of normal saline was injected in natural cycle group.After last injection,female and male mice at a ratio of 2∶1 were caged in each group.The vaginal plug,number of embryo implantation,histological changes of the endometrium during implantation window,and the number of spiral arteries in the endometrium were assessed on the next morning.Mice were sacrificed after mating day,with six mice per day since the 2 nd day.The expression of macrophage inflammatory protein 1β(MIP1β),CD244,and uterine natural killer cell(uNK)protein were detected.Results The number of embryo implantation in ovulation induction group significantly decreased on the 2 nd day after the mating day,while the number of spiral arteries in the endometrium decreased on the 4 th day compared to natural cycle group(P<0.05).Compared to ovulation induction group,there were significantly larger numbers of embryo implantation and spinal arteries in the endometrium in progesterone group and Modified Lianggu Decoction group(P<0.05).The expression of CD244 and uNK protein significantly decreased on the 4 th and 5 th day in ovulation induction group compared to the natural
